摘 要:以海藻酸钠为唯一碳源,从腐烂的海带中筛选得到褐藻胶裂解酶高产菌株YSH5,通过形态特征、生理生化特征、16S rDNA序列分析及系统发育树分析等对其进行鉴定,并采用DNS法研究了菌株YSH5所产褐藻胶裂解酶的酶学性质。结果表明,菌株YSH5为假单胞菌属(Pseudomonas sp.);液体发酵培养后菌株YSH5所产褐藻胶裂解酶的酶活最高达到58.66 U·mL^(-1);该酶的最适温度为40℃,在30~40℃范围内具有良好的热稳定性,最适pH值为7.5;Na^(+)对褐藻胶裂解酶的活性具有促进作用,Mn^(2+)和Zn^(2+)对褐藻胶裂解酶的活性没有显著影响,K^(+)、Mg^(2+)、Ca^(2+)、Cu^(2+)、Fe^(2+)对褐藻胶裂解酶的活性具有不同程度的抑制作用,其中Cu^(2+)的抑制作用最强;褐藻胶裂解酶降解海藻酸钠得到的主要产物为聚合度3~5的褐藻寡糖,该酶属于双功能内切酶。Using sodium alginate as the only carbon source,we screened high-yield alginate lyase-producing strain YSH5 from rotten kelp,and identified the strain by morphological characteristics,physiological and biochemical characteristics,16S rDNA sequence analysis,and phylogenetic tree analysis.Moreover,we studied the enzymatic properties of alginate lyase produced by strain YSH5 through DNS method.The results show that strain YSH5 is identified as Pseudomonas sp..After liquid fermentation,the activity of alginate lyase produced by strain YSH5 reaches the highest of 58.66 U·mL^(-1).The optimum temperature of alginate lyase produced by strain YSH5 is 40℃,which has good thermal stability in the range of 30-40℃,and the optimum pH value is 7.5.Na^(+)can promote the activity of alginate lyase,Mn^(2+)and Zn^(2+)have no significant effect on the activity of alginate lyase,while K^(+),Mg^(2+),Ca^(2+),Cu^(2+),and Fe^(2+)have different inhibitory effects on the activity of alginate lyase,among which Cu^(2+) has the strongest inhibitory effect.The main product of sodium alginate degradation by alginate lyase is alginate oligosaccharides with polymerization degree of 3-5,which belongs to bifunctive endonuclide enzyme.
