摘  要:探究瓶尔小草的最佳采收时期,优化瓶尔小草总黄酮的提取工艺,并评价其体外抗氧化活性。以总黄酮含量为考察指标,对乙醇浓度、料液比、超声时间、提取温度进行单因素实验,并结合响应面法设计优选超声波辅助提取工艺。结果表明:瓶尔小草总黄酮的最优工艺条件为超声提取温度为40℃,乙醇浓度为65%,料液比为1∶30(g/mL),超声时间为35 min;一年中瓶尔小草的总黄酮含量变化存在两个富集期,最佳采收期为8月,其次4月份也较为适宜采收。以维生素C为对照,瓶尔小草黄酮提取物对DPPH自由基、ABTS^(+)自由基清除率分别可达94.76%和98.33%,瓶尔小草总黄酮的总还原能力测定IC_(50)为0.0157 mg/mL,说明瓶尔小草总黄酮具有良好的体外抗氧化活性。本研究为瓶尔小草的采收、开发利用提供了试验依据。The optimal harvesting period of Ophioglossum vulgatum L.was explored,the extraction process for total flavonoids of Ophioglossum vulgatum L.was optimized,and its antioxidant activity in vitro was evaluated.a single-factor experiment examined the effect of ethanol concentration,solid-liquid ratio,ultrasonic time,and extraction temperature.Response surface methodology was then used to optimize the ultrasonic-assisted extraction process.Results revealed the optimal extraction conditions for total flavonoids of Ophioglossum vulgatum L.were as follows:extraction temperature of 40℃,ethanol concentration of 65%,solid-to-liquid ratio of 1∶30(g/mL),and ultrasonic time of 35 minutes.There were two enrichment periods in the total flavonoid content of Ophioglossum vulgatum L.during the year,with the best harvesting period in August and the second-best in April.Compared to vitamin C,O.vulgatum L.flavonoids possessed strong in vitro activity,with DPPH and ABTS radical scavenging rates reaching 94.76%and 98.33%,respectively;the IC_(50) for total flavonoids of Ophioglossum vulgatum L.was 0.0157 mg/mL.This study provides a reference for harvesting and utilization of Ophioglossum vulgatum L.
