摘 要:为了探究固态发酵果胶酶的酶蛋白组成和活力、体外酶解豆粕效果及对蛋鸡生产性能的影响,试验采用液相色谱串联质谱技术对果胶酶进行酶蛋白组成分析;采用现行国家标准或者行业标准对果胶酶酶系中不同种类酶进行酶活力的测定;采用体外模拟胃-肠两步消化法,以豆粕为酶解底物,在反应体系中添加50 U/g的果胶酶消化5 h,研究果胶酶对豆粕的酶解效果;在蛋鸡日粮中添加100 mg/kg果胶酶(50000 U/g),饲喂35 d,研究果胶酶对蛋鸡生产性能的影响。结果表明:固态发酵果胶酶酶系中共检测出51种酶蛋白,包括果胶酶类14种,木聚糖酶类8种,蛋白酶类6种,纤维素酶类9种,β-甘露聚糖酶类2种,阿魏酸酯酶类2种,淀粉酶类4种,半乳糖苷酶类3种,其他酶类3种。固态发酵果胶酶酶系中果胶酶活力为50794 U/g,木聚糖酶活力为1521 U/g,β-葡聚糖酶活力为3942 U/g,糖化酶活力为4712 U/g,低温淀粉酶活力为230 U/g,酸性蛋白酶活力为198 U/g,中性蛋白酶活力为102 U/g,β-甘露聚糖酶活力为87 U/g,纤维素酶活力54 U/g。果胶酶组酶解豆粕产物中半乳糖醛酸的含量为78.42 mg,比对照组提高了25.01%(P<0.05);干物质消失率为66.60%,比对照组提高了4.29%(P<0.05)。果胶酶组料蛋比比对照组降低了3.06%(P<0.05),每只鸡比对照组多增加收入约0.008元/d。说明固态发酵果胶酶的酶系丰富,对豆粕具有较好的酶解效果;在蛋鸡日粮中添加100 mg/kg的固态发酵果胶酶,能够提高蛋鸡的经济效益,具有较高的市场应用潜力。In order to investigate the enzyme protein composition and activity of solid fermentation pectinase,the effects of enzymatic hydrolysis of soybean meal in vitro and its effect on the production performance of laying hens,the enzyme protein composition of pectinase was identified by liquid chromatography-tandem mass spectrometry.The activity of different kinds of enzymes in pectinase system was measured by the current national or industrial standard.Using soybean meal as substrate,50 U/g pectinase was added into the reaction system for 5 h to study the enzymatic hydrolysis effects of pectinase on soybean meal by simulated gastroenteric digestion in vitro.The hens were fed a diet supplemented with 100 mg/kg pectinase(50000 U/g)for 35 days to study the effects of pectinase on performance of laying hens.The results showed that a total of 51 different types of enzymes with different enzymolysis ways were detected in the pectinase system produced by solid-state fermentation,including 14 pectinases,8 xylanases,6 proteases,9 cellulases,2β-mannanases,2 ferulic acid esterases,4 amylases,3 galactosidases,and 3 other enzymes.In solid-state fermentation pectinase enzyme system,pectinase activity was 50794 U/g;xylanase activity was 1521 U/g;β-glucanase activity was 3942 U/g;glucoamylase activity was 4712 U/g;low-temperature amylase activity was 230 U/g;acidic protease activity was 198 U/g;neutral protease activity was 102 U/g;β-mannanase activity was 87 U/g,and cellulase activity was 54 U/g.The content of galacturonic acid in the enzymatic hydrolysates of the pectinase group was 78.42 mg while the control group was 62.73 mg,so the content of galacturonic acid increased by 25.01%(P<0.05),and the disappearance rate of dry matter in the pectinase group was 66.60%while the control group was 63.86%,so the disappearance rate of dry matter increased by 4.29%(P<0.05);the feed/egg ratio of the pectinase group decreased by 3.06%compared to the control group(P<0.05),and the income of each chicken in the pectinase group increased by 0.
