摘 要:目的:探讨番茄红素对麦芽酚铝诱导的海马损伤的可能保护作用。方法:采用麦芽酚铝腹腔注射的方法构建记忆障碍阿尔茨海默病大鼠模型,番茄红素灌胃作为干预措施。通过行为学实验Morris水迷宫实验评估SD大鼠空间记忆和学习能力是否受损,通过对海马组织进行炎症因子、APP和Tau的测定进而判断番茄红素的效果。结果:与对照组相比,铝暴露组大鼠学习记忆能力降低(P<0.05),海马组织中IL-1β、IL-6、TNF-α等炎症因子水平高于对照组(P<0.05)。与铝暴露组相比,番茄红素能改善SD大鼠的空间记忆和学习能力(P<0.05),降低IL-1β、IL-6、TNF-α等炎症因子的表达水平(P<0.05)。各组大鼠体内其他金属元素如Mg、Fe、Cu、Zn、Se等含量存在差异,可能与麦芽酚铝染毒或者番茄红素干预相关。结论:铝暴露会损伤SD大鼠空间记忆和学习能力,且能影响Mg、Fe、Cu、Zn、Se等金属代谢。番茄红素能够改善铝对SD大鼠空间记忆和学习能力的损伤,其机制可能与缓解大鼠海马神经炎症损伤有关;还可能改善铝暴露对其他金属元素含量产生的影响。Objective:To investigate the possible protective effect of lycopene on maltol aluminum-induced hippocampal lesions.Methods:A rat model of Alzheimer’s disease with memory impairment was constructed by intraperitoneal injection of maltol aluminum,and lycopene gavage was used as an intervention.The Morris water maze experiment was used to evaluate whether the spatial memory and learning ability of SD rats were impaired,and the inflammatory factors,APP and Tau were measured in hippocampal tissue.Results:Compared with the control group,the learning and memory ability of the aluminum-exposed rats was reduced(P<0.05),and the levels of inflammatory factors such as IL-1β,IL-6 and TNF-αin the hippocampus were significantly higher than those in the control group(P<0.05).Compared with the aluminum-exposed group,lycopene could improve the spatial memory and learning ability of SD rats(P<0.05),and reduce the levels of IL-1β,IL-6,TNF-αand other inflammatory factors(P<0.05).There were differences in the contents of other metal elements such as Mg,Fe,Cu,Zn and Se in the rats in each group,which may be related to the toxicity of aluminum maltol or the intervention of lycopene.Conclusion:Aluminum exposure impaired spatial memory and learning ability in SD rats,and could affect the metabolism of metals such as Mg,Fe,Cu,Zn,and Se.Lycopene can improve the damage of aluminum on spatial memory and learning ability in SD rats,and its mechanism may be related to the alleviation of neuroinflammatory damage in the hippocampus in rats,and may also improve the effect of aluminum exposure on the content of other metal elements.
