摘  要:以沃柑皮和麸皮作为液体发酵碳源,利用实验室筛选的高产纤维素酶草酸青霉菌株UNN1产植物多糖降解酶。以滤纸酶、果胶酶活力为主要评价指标,分析发酵液中碳源配比、氮源、发酵液pH、发酵温度和发酵时间对酶活力的影响,并在单因素试验的基础上,通过正交试验优化液体发酵条件。结果表明:产滤纸酶和果胶酶的最佳液体发酵条件为沃柑皮:麸皮质量比1∶4、氮源为4 g/L蛋白胨、发酵液pH 5.5、发酵温度28℃(转速为180 r/min)、发酵时间96 h,在此条件下的滤纸酶活力为(0.308±0.008)U/mL、果胶酶活力为(3.892±0.028)U/mL。Citrus peel and wheat bran were used as liquid fermentation carbon sources to produce plant polysaccharide-degrading enzymes using laboratory-screened high cellulase-producing Penicillium oxalicum strain UNN1.The effects of carbon source ratio of fermentation broth,nitrogen source,pH of fermentation broth,fermentation temperature and fermentation time on enzyme activity were analyzed using filter paper cellulase,and pectinase activity as the main evaluation indexes,and the optimization of the liquid fermentation conditions was carried out by orthogonal test on the basis of single factor experiments.The results showed that the optimal liquid fermentation conditions for the production of filter paper cellulase and pectinase were citrus peel:wheat bran mass ratio 1∶4,4 g/L peptone as nitrogen source,pH of fermentation broth 5.5,fermentation temperature 28℃(rotational speed 180 r/min),fermentation time 96 h.Under the conditions,the filter paper cellulase activity was(0.308±0.008)U/mL and pectinase activity was(3.892±0.028)U/mL.
