金耳菌株的筛选与分离鉴定

摘  要:以7株金耳菌株为试材,采用不同菌株在cPDA固体平板培养基菌丝生长情况比较,并通过栽培试验进行比较筛选的方法,研究了不同菌株的生长情况,并对生产菌株进行分离及分子鉴定,以期为金耳栽培过程中优良生产菌株的选择提供参考依据。结果表明:7株生产菌株依据cPDA平板培养基生长情况,可以看出“金耳1号”“金耳2号”“金耳5号”及“金耳7号”的菌丝生长速率较高,栽培试验结果表明,7株金耳菌株均可出耳,但“金耳1号”的综合出耳效果最好,初步确认“金耳1号”可以作为生产菌株进行使用,将“金耳1号”通过孢子分离及基内菌丝分离法分离出了2种真菌,通过ITS测序及进化关系分析确定了2种真菌分别为金耳菌及层叠韧革菌。Taking 7 Naematelia aurantialba strains as the test materials,the mycelium growth of different strains in cPDA solid plate medium was compared,and the growth of different strains was studied through the cultivation test,and the production strains were separated and molecular identification,in order to provide reference for the selection of excellent strains during the cultivation of Naematelia aurantialba.The results showed that,according to the growth of the 7 production strains,it could be seen that the mycelium growth rate of‘Naematelia aurantialba 1’‘Naematelia aurantialba 2’‘Naematelia aurantialba 5’and‘Naematelia aurantialba 7’had higher mycelium growth rate.The cultivation test results showed that all the 7 strains of Naematelia aurantialba could produce ear,but‘Naematelia aurantialba 1’had the best comprehensive ear growth effect.It was preliminarily confirmed that‘Naematelia aurantialba 1’could be used as a production strain.Two fungi were isolated from‘Naematelia aurantialba 1’by spore separation and intrabasal mycelium separation.The two fungi were identified as Naematelia aurantialba and Stereum complicatum by ITS sequencing and evolutionary relationship analysis.
