摘  要:1-脱氧-D-木酮糖-5-磷酸合成酶(DXS)是MEP途径的第一个限速酶,在茶树抗性的形成中起到重要作用.为探究茶树DXS基因家族的潜在功能,利用生物信息学和实时荧光定量PCR技术,对茶树DXS基因家族进行鉴定及表达分析.采用生物信息学方法,从‘黄棪’和‘铁观音’茶树中分别鉴定出5个DXS基因家族成员,分别命名为HD-CsDXS1-CsDXS5和TGY-CsDXS1-CsDXS5,系统发育结果表明其分属3个亚家族,相同亚家族的CsDXS基因外显子-内含子数目及排列顺序基本一致.CsDXS启动子上存在众多与植物发育、激素和胁迫响应相关的顺式作用元件.荧光定量检测发现,HD-CsDXS2和HD-CsDXS5在MeJA、GA、ABA、低温和干旱处理下表达量均显著上调,HD-CsDXS1和HD-CsDXS4在GA、ABA和干旱处理下表达量显著上调,HD-CsDXS3仅在GA和干旱处理下表达量上调.本研究表明CsDXS在非生物胁迫下均被诱导表达,可能与茶树抗逆性密切相关,结果可为进一步探究茶树DXS基因家族的功能研究提供参考.(图8表2参30)1-Deoxy-D-xylulose-5-phosphate synthetase(DXS)is the first rate-limiting enzyme in the methylerythritol-4-phosphate(MEP)pathway of plant terpene synthesis,playing a crucial role in tea plant resistance.To investigate the potential functions of the DXS gene family,we employed bioinformatics analysis and quantitative reverse transcription PCR(qRT-PCR)to identify and analyze the expression patterns of the CsDXS gene family.Five DXS gene family members were identified from Huangdan and five from Tieguanyin tea plants,named HD-CsDXS1–5 and TGY-CsDXS1-5,respectively.Phylogenetic analysis categorized them into three subfamilies,with consistent exon-intron structure within each subfamily.Promoter analysis revealed numerous cis-acting elements associated with stress responses,hormones,and growth.Quantitative fluorescence detection indicated significant upregulation of HD-CsDXS2 and HD-CsDXS5 under methyl jasmonate(MeJA),gibberellin(GA),abscisic acid(ABA),low temperature,and drought treatments;HD-CsDXS1 and HD-CsDXS4 were significantly upregulated under GA,ABA,and drought treatments;and HD-CsDXS3 was upregulated only under GA and drought treatments.These findings suggest that CsDXS genes are induced under abiotic stress and may contribute to the stress resistance of tea plants.Our results provide a foundation for further research on the functions of the DXS gene family in tea plants.
