摘  要:旨在从蛋白水平揭示鹿皮胶和阿胶的蛋白质组分信息,探究鹿皮胶和阿胶的差异关键蛋白和代谢通路。以鹿皮胶和阿胶为试验材料,利用4D label-free定量蛋白质组学进行质谱鉴定,通过生物信息学技术分析蛋白的亚细胞定位、结构域、GO注释和KEGG通路注释。采用4D-label-free技术对鹿皮胶与阿胶蛋白组分进行了比较研究,得到有效图谱12 451个,鉴定肽段1 449,鉴定unique肽段1 035,鉴定蛋白数455,定量蛋白数303。上调差异蛋白主要集中在原发免疫缺陷、TGF信号通路、糖尿病并发症等功能中,下调差异蛋白主要集中在蛋白质的消化和吸收、松弛素信号通路、ECM受体相互作用、丙酮酸代谢物、非洲锥虫病、PI3K-AKT信号通路等功能中。鹿胶与阿胶进行差异蛋白统计比较,下调68个,上调56个,主要参与了蛋白质合成、发育、吸附、细胞骨架、转运等生物学过程。The purpose of this paper is to reveal the information of protein components of deer skin glue and ejiao from the protein level,and to explore the differences of key proteins and metabolic pathways between deer skin glue and Donkey-hide glue.The protein subcellular location,domain,GO annotation and KEGG pathway annotation were analyzed by bioinformatics using 4D label-free quantitative proteomics.The results show that:The 4D-label-free technique was used to compare the components of deer skin glue and ejiao protein.12451 effective maps were obtained,including 1449 identified peptides,1035 unique peptides,455 identified proteins,and 303 quantitative proteins.Up-regulated differential proteins are mainly concentrated in primary immunodeficiency,TGF signal pathway and diabetic complications,while downregulated differential proteins are mainly concentrated in protein digestion and absorption,relaxin signal pathway,ECM receptor interaction,pyruvate metabolites,African trypanosomiasis,PI3K-AKT signal pathway and so on.There were 68 down-regulated proteins and 56 up-regulated proteins between deer gelatin and ejiao.Mainly involved in protein synthesis,development,adsorption,cytoskeleton,transport and other biological processes.
