摘  要:试验利用生物化学、药理学及分子生物学方法提取戊糖片球菌抗菌肽粗提物,测定其大小及抑菌性,并将其配制成制剂,按高、中、低3种剂量饲喂察哈尔羊,以研究其对察哈尔羊免疫性能及肠道的微生物多样性的影响。结果显示:抗菌肽粗提物大小约为35 KD,对金黄色葡萄球菌、沙门氏菌及大肠杆菌均具有极显著的抑制效果(P<0.01),且IgA,IgM及IgG相对于对照组浓度均显著提高(P<0.05);厚壁菌门和拟杆菌门为微生物群落最具优势的门,且高剂量试验组细菌微生物群落丰富度最高。功能预测抗菌肽促进了碳水化合物代谢、跨膜运输和氨基酸代谢。这些研究为后期戊糖片球菌抗菌肽的研究提供理论依据。The study used biochemical,pharmacological,and molecular biology methods to extract the crude extract of antimicrobial peptides from Pediococcus pentosaceus,and measured its size and antibacterial activity.At the same time,Chahar sheep were fed to study its immune performance and intestinal microbial diversity.The results showed that the size of the crude extract of antimicrobial peptides was approximately 35 KD;it had extremely significant inhibitory effects on Staphylococcus aureus,Salmonella,and Escherichia coli(P<0.01);and the concentrations of IgA,IgM,and IgG increased compared to the blank control group(P<0.05);firmicutes and Bacteroidetes were the most dominant phyla in bacterial microbial communities,and the high-dose experimental group had the highest richness of bacterial microbial communities.Functional predictions suggest that antimicrobial peptides promote carbohydrate metabolism,transmembrane transport,and amino acid metabolism.These studies provided a theoretical basis for the study of antimicrobial peptides against Pediococcus pentosaceus in the later stage.
