摘  要:传统的基于球状胶体金(Au spheres,AuSPs)试纸条由于检测灵敏度偏低,无法满足食品安全高灵敏检测的需求。因此,建立高灵敏、快速分析方法尤为重要。以黄曲霉毒素B_(1)(aflatoxin B_(1),AFB_(1))为检测对象,金纳米棒(Au nanorods,AuNRs)作为信号标签,偶联抗AFB_(1)单克隆抗体作为金标探针,基于竞争法制备免疫层析试纸条对玉米中AFB_(1)进行检测,试验结果通过便携式试纸条定量分析仪定量检测。结果表明:在单抗加入量为0.6μg、T线抗原质量浓度为0.1 mg/mL、探针使用体积为2.5μL的条件下制作标准曲线,基于AuNRs的试纸条检出限为0.124 ng/mL,IC50为1.593 ng/mL,此外实际玉米样品中对AFB_(1)的回收率为89.50%~111.70%;对玉米质控样进行检测,结果与所建立的方法一致。该方法可应用于玉米中AFB_(1)的快速检测,检测结果符合国家标准,具有良好的应用前景。Traditional Au spheres(AuSPs)based test strips cannot meet the requirements of high-sensitive detection of food safety due to their low detection sensitivity.Therefore,it is particularly important to establish highly sensitive and rapid analysis methods.With aflatoxin B_(1)(aflatoxin B_(1),AFB_(1))as the detection object,Au nanorods(AuNRs)as the signal tag,conjugated anti-AFB_(1) monoclonal antibody as the gold labeled probe,immunochromatographic test strip were prepared based on competition method for sensitive detection of AFB_(1) in maize.The experimental results were quantified by a portable test strip quantitative analyzer.The standard curve was prepared under the conditions of 0.6μg monoclonal antibody addition,0.1 mg/mL T-line antigen concentration and 2.5μL probe usage volume,the detection limit of the strip based on AuNRs was 0.124 ng/mL,IC 50 was 1.593 ng/mL.In addition,the recovery rate of AFB_(1) in the actual maize samples was 89.5% to 111.70%.The maize quality control samples were tested,and the results were consistent with the established method.The method can be applied to the rapid detection of AFB_(1) in maize,and the sample detection results meet the national standard and have a good application prospect.
