摘 要:为探究酸枣仁-藤茶酒对小鼠睡眠的改善作用,通过腹腔注射对氯苯丙氨酸(PCPA)构建失眠小鼠模型。实验一组小鼠进行睡眠诱导实验,观察其睡眠潜伏期和睡眠时间;实验二组小鼠进行旷场实验后检测小鼠脑内相关神经递质含量。结果与模型组相比,酸枣仁-藤茶酒能够显著缩短实验小鼠的睡眠潜伏期,延长小鼠睡眠时间(P<0.05,P<0.01),抑制小鼠的自主活动;能够增加小鼠海马体中γ-氨基丁酸(GABA)含量并减少谷氨酸(Glu)的含量(P<0.05),维持受试小鼠海马体中Glu/GABA比值的平衡;酸枣仁-藤茶酒高剂量组(80 g/kg)能够显著增加小鼠下丘脑中5-羟色胺(5-HT)以及其代谢产物5-羟基吲哚乙酸(5-HIAA)的含量(P<0.01),并减少小鼠下丘脑中去甲肾上腺素(NE)的分泌。实验表明酸枣仁-藤茶酒表现出明显的抗焦虑和镇静催眠活性,其作用机制与调控脑内睡眠相关的神经递质合成与释放有关。To investigate the sleep-improving effects of sour jujube kernel-Teng cha wine on mice with insomnia,insomnia mice models were created by intraperitoneal injection of p-chlorophenylalanine(PCPA).The first group of mice underwent sleep induction experiments to observe sleep latency and sleep duration,while the second group of mice underwent open field tests followed by detection of neurotransmitter levels in the brain.Compared to the model group,sour jujube kernel-Tengcha wine significantly reduced the sleep latency and prolonged sleep duration of the experimental mice(P<0.05,P<0.01),suppressed their spontaneous activity,increased the gamma-aminobutyric acid(GABA)content in the hippocampus and reduced the glutamate(Glu)content(P<0.05),maintaining a balance in the Glu/GABA ratio in the hippocampus of the tested mice,and suppressed spontaneous activity.The high-dose group of sour jujube kernel-Tengcha wine(80 g/kg)significantly increased the content of serotonin(5-HT)and its metabolite 5-hydroxyindoleacetic acid(5-HIAA)in the hypothalamus of mice(P<0.01),and reduced the secretion of norepinephrine(NE)in the hypothalamus of mice.These findings indicated that sour jujube seed-Tengcha wine exhibited significant anti-anxiety and sedative-hypnotic activities,and its mechanism of action was related to the regulation of neurotransmitter synthesis and release in the brain related to sleep.
