摘  要:从野生紫芝子实体5个不同部位分离纯培养获得其菌丝体,测试6种培养基的菌丝体培养效果,通过单因素试验和正交试验对紫芝菌丝体液体发酵工艺条件进行优化,测试菌丝体在系列化添加H_(2)SeO_(3)的培养基中1~6 d的硒含量以考察其富硒动力学。结果表明:紫芝菌盖边缘部位的离体组织无性生殖能力最强;初筛试验显示由1.5%玉米浸提液和2%的蔗糖组合而成的玉米蔗糖培养基最佳;正交试验所得最佳发酵工艺条件为发酵温度为25℃、振荡速度为140 r/min、培养基装液量为80 mL;复筛试验所得最佳营养素搭配为:玉米蔗糖培养基、0.025%MgSO_(4)、0.15%KH_(2)PO_(4)、0.3%蛋白胨,优化后紫芝菌丝体增产48.96%;菌丝体硒含量随培养基中硒浓度的升高而提高,其富硒动力学过程符合指数增长模型,菌丝体的硒含量≥0.24μg/g。此研究开发了东北寒地黑土区特有的真菌资源和玉米资源,为借助真菌菌丝体生产富硒食品原料提供了一条新途径。The mycelium tissues were isolated and cultivated from 5 different parts of wild fruiting bodies of Ganoderma sinense,The cultivation efficiency of mycelium tissues on six different media was tested.Liquid fermentation process of Ganoderma sinense mycelium was optimized using single factor experiments and orthogonal experiments.Mycelium selenium enrichment kinetics was assessed in a series of H_(2)SeO_(3)-supplemented fermentation media over 1 to 6 days.The results showed that detached tissues from the edge of Ganoderma sinense exhibited the strongest asexual reproductive ability.Initial screening tests indicated that a combination of 1.5%corn extract liquor and 2%sucrose formed the optimal corn-sucrose medium.Orthogonal experiments showed optimal fermentation conditions were as follows:a fermentation temperature of 25℃,an oscillation speed of 140 r/min,and a liquid volume of 80 mL in the medium.The optimal nutrient medium obtained by secondary screening tests was corn-sucrose medium with 0.025%MgSO_(4),0.15%KH_(2)PO_(4)and 0.3%peptone,resulting in a 48.96%yield increase in Ganoderma sinense mycelium production post-optimization.The selenium content of mycelium increased with the increase in selenium concentration in the medium,following an exponential growth model during the selenium-enriched process,with mycelium selenium content≥0.24μg/g.This study developed unique fungal and corn resources from Northeast China black soil region,providing a new approach for producing selenium-enriched food ingredients through fungal mycelium cultivation.
