提取温度对香菇多糖结构及生物活性的影响

摘  要:本研究以香菇子实体为原料,采用HPLC、FT-IR、AFM、SEM法检测100、121和133℃温度下提取的香菇多糖的结构与成分的差异;并采用吞噬中性红、CCK-8法检测香菇多糖体外生物活性。结果显示,不同温度下提取的三种多糖均由β-葡萄糖聚合而成,133℃提取的香菇多糖分子量较100和121℃提取的香菇多糖分子量增大了10倍。体外免疫活性研究显示,香菇多糖对三种细胞的影响按大小依次为RAW264.7细胞吞噬能力>T(Jurkat)细胞增殖>B(Raji)细胞增殖。100和121℃提取的高分子量香菇多糖对RAW264.7细胞吞噬能力和T(Jurkat)细胞增殖均有显著促进作用(P<0.05)。本研究表明香菇多糖的最适提取温度为100~121℃,香菇多糖中主要活性成分的分子量大小为200~6000 kDa。该研究结果为香菇多糖提取工艺的优化提供了一定的科学依据。In this study,Lentinus edodes fruiting bodies was used as raw material,the effects of extraction temperature(100,121,and 133℃)on the structure,composition,and bioactivity of polysaccharides extracted from Lentinus edodes fruiting bodies were investigated.The extracted polysaccharides were characterized by HPLC,FT-IR,AFM and SEM.The in vitro biological activity of Lentinus edodes polysaccharides was assessed using phagocytic neutral red and CCK-8 assay.All three polysaccharides extracted under different temperature were found to beβ-glucans.The polysaccharide extracted at 133℃exhibited a 10-fold higher molecular weight compared to those extracted at 100 and 121℃.In vitro immunoreactivity study showed that the effects of Lentinus edodes polysaccharides on the three cell types,in descending order,were:RAW 264.7 cell phagocytosis>T(Jurkat)cell proliferation>B(Raji)cell proliferation.Polysaccharides extracted at 100 and 121℃significantly promoted phagocytosis of RAW 264.7 cells and proliferation of T(Jurkat)cells(P<0.05).Therefore,the optimum extraction temperature of Lentinus edodes polysaccharides was 100〜121℃,and the molecular weight of the main active components in Lentinus edodes polysaccharides was determined to be 200〜6000 kDa.These findings provide a scientific basis for the optimization of the extraction process of Lentinus edodes polysaccharides.
