摘  要:目的:探讨蛇葡萄素(AMP)调控过氧化物酶体增殖物激活受体γ(PPARγ)信号通路对骨肉瘤细胞生物学行为的影响。方法:取生长良好的U2OS细胞进行分组:空白组、20μmol/L AMP组、40μmol/L AMP组、80μmol/L AMP组、80μmol/L AMP+GW9662(PPARγ抑制剂)组、80μmol/L AMP+ROZ(PPARγ激活剂-罗格列酮)组。CCK-8法及克隆形成实验检测细胞增殖;Transwell实验及划痕实验检测U2OS细胞侵袭与迁移;流式细胞术检测U2OS细胞凋亡;Western blot检测Bcl-2、PCNA、MMP-9、PPARγ蛋白表达水平。结果:与空白组相比,20μmol/L AMP组、40μmol/L AMP组、80μmol/L AMP组U2OS细胞增殖、克隆数、侵袭、迁移率、Bcl-2、PCNA、MMP-9表达显著降低,凋亡率、PPARγ表达显著增加(P<0.05);与80μmol/L AMP组相比,80μmol/L AMP+GW9662组U2OS细胞增殖、克隆数、侵袭、迁移率、Bcl-2、PCNA、MMP-9表达显著增加,凋亡率、PPARγ表达显著降低(P<0.05),但80μmol/L AMP+ROZ组U2OS细胞增殖、克隆数、侵袭、迁移率、Bcl-2、PCNA、MMP-9表达显著降低,凋亡率、PPARγ表达显著增加(P<0.05)。结论:AMP通过激活PPARγ信号通路抑制骨肉瘤细胞恶性生物学行为。Objective:To investigate the effect of ampelopsin(AMP)on the biological behavior of osteosarcoma cells by regulating the peroxisome proliferator-activated receptorγ(PPARγ)signaling pathway.Methods:U2OS cells with good growth were separated into six groups:Blank group,20μmol/L AMP group,40μmol/L AMP group,80μmol/L AMP group,80μmol/L AMP+GW9662(PPARγinhibitor)group,and 80μmol/L AMP+ROZ(PPARγactivator-rosiglitazone)group.CCK-8 method and clone formation experiment were applied to detect cell proliferation.Transwell and scratch experiments were applied to detect the invasion and migration of U2OS cells.Flow cytometry was applied to detect U2OS cell apoptosis.Western blot was applied to detect the expression levels of Bcl-2,PCNA,MMP-9,and PPARγproteins.Results:Compared with the blank group,the proliferation,clone number,invasion,migration rates,Bcl-2,PCNA,and MMP-9 expression of U2OS cells in the 20μmol/L AMP group,40μmol/L AMP group,and 80μmol/L AMP group obviously reduced,the apoptosis rate and PPARγexpression obviously increased(P<0.05).Compared with the 80μmol/L AMP group,the proliferation,clone number,invasion,migration rates,Bcl-2,PCNA,and MMP-9 expression of U2OS cells in the 80μmol/L AMP+GW9662 group obviously increased,the apoptosis rate and PPARγexpression significantly reduced(P<0.05).However,the proliferation,clone number,invasion,migration rates,Bcl-2,PCNA,and MMP-9 expression of U2OS cells in the 80μmol/L AMP+ROZ group significantly reduced,the apoptosis rate and PPARγexpression obviously increased(P<0.05).Conclusion:AMP inhibits the malignant biological behavior of osteosarcoma cells by activating the PPARγsignaling pathway.
