猪卵丘细胞和壁颗粒细胞转录组差异分析

摘  要:伴随着卵泡腔的形成,颗粒细胞逐步分化成两种不同类型:壁颗粒细胞(mural granulosa cells,MGCs)和卵丘细胞(cumulus cells,CCs)。目前许多研究表明壁颗粒细胞和卵丘细胞存在功能差异。壁颗粒细胞和卵丘细胞在卵母细胞生长发育过程中发挥着非常重要的作用,在细胞周期、DNA复制、缝隙连接等重要环节担任重要角色。为研究壁颗粒细胞和卵丘细胞存在的功能差异,我们利用转录组测序技术分析了壁颗粒细胞和卵丘细胞的差异基因,又通过GO分析和KEGG信号通路分析对差异基因进行富集。结果表明,壁颗粒细胞和卵丘细胞转录组存在较大差异,两者差异显著基因多达3305个,与壁颗粒细胞相比,卵丘细胞在细胞周期和DNA复制信号通路中大多数基因表达下调,而在缝隙连接信号通路中大多数基因表达上调,两者功能上存在较大差异。Along with the formation of the follicular lumen,granulosa cells gradually differentiate into two different types:cumulus cells and mural granulosa cells.Many studies have shown that there are functional differences between cumulus cells and mural granulosa cells.Cumulus cells and mural granulosa cells play a very important role in the development of oocytes and play an important role in the cell cycle,DNA replication,and gap junctions.To investigate the functional differences between cumulus cells and mural granulosa cells,we analyzed the differential genes between cumulus cells and mural granulosa cells using transcriptome sequencing technology,and enriched the differential genes by GO analysis and KEGG signaling pathway analysis.The results showed that the transcriptomes of cumulus cells and mural granulosa cells were very different,with as many as 3305 significantly different genes,and the expression of most genes in the cell cycle and DNA replication signaling pathways was down-regulated in cumulus cells compared with mural granulosa cells,while the expression of most genes in the gap junction signaling pathway was up-regulated,and there were large functional differences.
