摘 要:该研究基于一种三流体同轴喷雾干燥技术,以明胶和结冷胶为壁材,以CaCl 2为交联剂,通过凝聚、离子交联或者组合将植物乳杆菌包埋于微胶囊中。对微胶囊的微观结构、溶解性、傅里叶红外光谱进行分析,并测定了益生菌微胶囊的喷雾干燥存活率,模拟消化稳定性和贮藏稳定性。结果显示,益生菌被成功包埋于微胶囊内部;结冷胶及Ca^(2+)的添加显著降低了明胶微胶的溶解性和溶胀性,傅里叶红外光谱结果证实了明胶和结冷胶间的氢键和静电相互作用以及Ca^(2+)和羧基间的交联作用。明胶-结冷胶-钙离子微胶囊在喷雾干燥、模拟胃液消化、4℃贮藏8周后菌活分别为9.71、8.74和9.47 lg CFU/g。综上所述,Ca^(2+)交联的明胶-结冷胶复合微胶囊可以为益生菌在胁迫环境中提供良好的保护。This study is based on a three-fluid concentric spraying drying technology,using gelatin and gellan gum as wall materials and calcium chloride as crosslinking agent,to encapsulate Lactobacillus plantarum in microcapsules through coacervation,calcium ion cross-linking,or a combination of both.The microcapsules were analyzed using freeze-scanning electron microscopy,solubility tests,and Fourier-transform infrared spectroscopy.The spray-drying survival rate,simulated digestion stability,and storage stability of the probiotic microcapsules were also measured.Results showed that the probiotics were successfully encapsulated within the microcapsules.The addition of gellan gum and calcium ions significantly reduced the solubility and swelling capacity of the gelatin microcapsules.Fourier-transform infrared spectroscopy confirmed the hydrogen bonds and electrostatic interactions between gelatin and pectin,as well as the cross-linking between calcium ions and carboxyl groups.After spray drying,simulated gastric fluid digestion,and 8 weeks of storage at 4℃,the microbial activity was 9.71,8.74,and 9.47 lg CFU/g,respectively.In conclusion,the calcium ion-crosslinked gelatin-gellan gum composite microcapsules could provide excellent protection for probiotics in harsh environments.
