摘  要:目的:建立一种快速、高效的鸡精调味料中鸡源性成分的提取和分析方法,并应用于鸡精调味料真伪鉴别的风险监测。方法:对比CTAB法、磁珠法和硅胶柱法提取鸡精调味料中DNA,通过后期实时荧光PCR反应确定DNA的提取效果,建立一整套完善的鸡精调味料中鸡源性成分的检测方法,并基于建立的方法进行鸡精调味料的风险监测。结果:相比CTAB法,磁珠法和硅胶柱法的提取效果更好,更适合用于鸡精调味料的DNA提取。磁珠法在高通量、快速、操作简便方面优于硅胶柱法,适合样品的大批量检测,但在DNA破坏较为严重时,建议结合硅胶柱法对扩增结果进一步确认,以防误判。基于建立的方法对鸡精调味料产品进行风险监测,30批次样品发现2批次未检出鸡源性成分,问题发现率为6.67%。结论:基于磁珠法建立了高通量、快速及简便的鸡精调味料中鸡源性成分的检测方法,为政府监管部门的风险监测提供了技术支持。Objective:To establish an rapid and efficient method for the extraction and analysis of chicken derived material in chicken essence seasoning,and apply to study the risk monitoring for the authenticity identification of chicken essence seasoning.Methods:Compared the CTAB method,magnetic bead method and silica gel column method to extract DNA from chicken essence seasoning,the effect of DNA extraction was determined by real-time PCR.A complete detection method for chicken derived components in chicken essence seasoning was established and the risk monitoring of chicken essence seasoning was carried out based on the established methods.Results:Compared with CTAB method,magnetic bead method and silica gel column method had better extraction effect,and the latter two methods were more suitable for DNA extraction of chicken essence seasoning.Magnetic bead method is superior to silica gel column method in terms of high-throughput,rapid speed and simple operation,which is suitable for mass detection of samples.However,when DNA is seriously damaged,it is recommended to further confirm the amplification results with silica gel column method to prevent misjudgment.Based on the established method,the risk monitoring of chicken essence seasoning products was carried out.It was found that no chicken-derived components were detected in 2 batches of 30 samples,and the problem detection rate was 6.67%.Conclusion:Based on magnetic bead method,a high throughput,rapid and simple method for the detection of chicken-derived components in chicken essence seasoning was established,which provided technical support for the risk monitoring of government supervision departments.
