摘 要:对核桃青皮中胡桃醌含量检测中提取试剂及提取液中胡桃醌的稳定性进行研究。以核桃青皮提取液中胡桃醌含量为指标,考察不同体积分数乙酸的50%甲醇溶液提取核桃青皮提取液中胡桃醌含量变化情况;研究对比氯仿、氯仿-乙酸(1︰1,V/V)、乙酸-甲醇(1︰1,V/V)及乙酸-50%甲醇(1︰1,V/V)4种溶剂的核桃青皮提取液中胡桃醌的提取效果;对比验证氯仿-乙酸(1︰1,V/V)、氯仿-乙酸(2︰3,V/V)及氯仿-乙酸(3︰2,V/V)3种溶剂下,核桃青皮提取液中胡桃醌含量的稳定性;对超声时间进行试验验证。结果发现,使用氯仿-乙酸(1︰1,V/V)溶剂超声150min后的核桃青皮提取液中胡桃醌含量相对较高,且样品液中胡桃醌含量在240 min内稳定性良好,能够较好满足核桃青皮中胡桃醌含量检测的要求。试验得出一种较为稳定的核桃青皮中胡桃醌含量检测前处理方法,为相关领域的学术研究和工业应用提供参考。The stability of the extraction reagent and the extraction solution of juglone in walnut peel were studied.The content of juglan quinone in walnut peel extract was investigated by 50%methanol solution with different concentration of acetic acid.The extraction efficiency of juglone from walnut peel extract was compared with chloroform,chloroform-acetic acid(1︰1,V/V),acetic acid-methanol(1︰1,V/V)and acetic acid-50%methanol(1︰1,V/V).The stability of juglone content in three kinds of walnut peel extracts with chloroform-acetic acid(1︰1,V/V),chloroform-acetic acid(2︰3,V/V)and chloroform-acetic acid(3︰2,V/V)solvents was verified by comparison.The ultrasonic time was verified by experiment.The studies showed that using chloroform,the content of juglone in walnut peel extract was relatively high after 150 min ultrasonic extraction with acetic acid(1︰1,V/V)solvent,and the content of juglone in sample solution was stable within 240 min,which could better meet the requirements for the detection of juglone content in walnut peel.A relatively stable pretreatment method for the determination of juglone content in walnut peel was obtained,which provided reference for academic research and industrial application in related fields.
