摘 要:为了对TRPM1基因SNPs位点进行基因分型,分析多态位点与皮肤乌色度的关联性,找出对皮肤乌色度相关的SNP标记,采用基于飞行时间质谱分析技术的MassARRAY平台对TRPM1基因的SNPs位点进行基因分型,分析单个SNP标记与皮肤亮度L^(*)值性状的相关性,并分析不同基因型与皮肤亮度L^(*)值性状的相关性。TRPM1基因6个SNP位点质谱检出率为100%,只有SNP1位点(第8外显子,rs737038441)具有多态性,有3种基因型。卡方检验表明,该SNP位点处于Hardy-Weinberg平衡状态(P>0.05),PIC=0.27,为中度多态(0.25<PIC<0.5)。单标记关联分析表明,SNP1位点GG基因型的背部皮肤亮度L^(*)值显著低于AA和AG基因型(P<0.05),但该SNP位点不同基因型间大腿部皮肤和胸部皮肤亮度L^(*)值均差异不显著。筛选到TRPM1基因的1个多态性SNP1位点(rs737038441 A>G突变),该位点处于H-W平衡状态,SNP1位点与丝羽乌骨鸡背部皮肤亮度L^(*)值显著相关,GG基因型背部皮肤亮度L^(*)值显著低于AA和AG基因型(P<0.05),GG基因型可能是丝羽乌骨鸡背部皮肤乌色度的优势基因型,为利用分子标记辅助育种加快培育乌色度高乌骨鸡新品种提供理论参考。This study aimed to genotype the single nucleotide polymorphisms(SNPs)of the TRPM1 gene and analyze the association between these polymorphic sites and skin blackness to identify SNP markers that correlate with skin blackness.The SNPs loci in TRPM1 gene was genotyped using MassARRAY platform based on time-of-flight mass spectrometry analysis technology and the correlation between single SNP markers,and the L^(*)value trait of skin brightness was analyzed.The mass spectrometry detection rate of 6 SNP loci in TRPM1 gene was 100%,and only SNP1 locus(exon 8,rs737038441)had polymorphism,with 3 genotypes.The chi square test showed that the SNP1 locus was in a Hardy Weinberg equilibrium state(P>0.05),with a PIC of 0.27,indicating moderate polymorphism(0.25<PIC<0.5).Single marker association analysis showed that the back skin brightness L^(*)value of the GG genotype at the SNP1 locus was significantly lower than that of the AA and AG genotypes(P<0.05),while there was no significant difference in the brightness L^(*)value of the thigh and breast skin between different genotypes at SNP1 locus.One polymorphic SNP locus(rs737038441 A>G mutation)of the TRPM1 gene was identified,which was in an H-W equilibrium state.SNP1 locus of the TRPM1 gene was significantly correlated with the brightness L^(*)value of the back skin of Silky fowls.The GG genotype had a significantly lower brightness L^(*)value of the back skin than that of the AA and AG genotypes(P<0.05).The GG genotype may be the dominant genotype for the dark color of the back skin of Silky fowls.It will provide a theoretical reference for using molecular marker assisted breeding to accelerate the cultivation of new varieties of high blackness black bone chickens.
