摘　　要：目的探讨ZNF460在乳腺癌中的表达及其对MCF-7细胞增殖的影响。方法应用qRT-PCR和Western blot法检测MCF-7和MCF10A细胞中ZNF460 mRNA和蛋白的表达;免疫组织化学方法分析ZNF460的表达与乳腺癌临床病理特征的关系;将ZNF460 siRNA转染至MCF-7细胞后,XTT法检测细胞增殖、qRT-PCR和Western blot法分析TRIM28和c-Myc表达的变化。结果ZNF460 mRNA和蛋白表达水平在MCF-7细胞中均明显高于MCF10A细胞(均P<0.01),其表达随着患者年龄的降低(P<0.05)以及组织学分级(P<0.01)和TNM分期(P<0.05)的增高而增高;ZNF460的表达下调后,MCF-7细胞的增殖能力明显降低(P<0.01),TRIM28和c-Myc mRNA和蛋白的表达水平明显降低(P<0.05)。结论ZNF460在MCF-7细胞中高表达,下调ZNF460表达能明显抑制MCF-7细胞的增殖能力,ZNF460可能通过促进TRIM28-c-Myc轴的激活参与乳腺癌的恶性进展。Objective To investigate the expression of ZNF460 in breast cancer and its effect on the proliferation of MCF-7 cells.Methods The expressions of ZNF460 mRNA and protein in MCF-7 and MCF10A cells were detected by qRT-PCR and Western blot.The relationship between ZNF460 expression and clinicopathological features of breast cancer was analyzed by immunohistochemistry.After ZNF460 siRNA was transfected into MCF-7 cells,the cell proliferation was detected by XTT method,and the expression of TRIM28 and c-Myc was analyzed by qRT-PCR and Western blot.Results The expression of ZNF460 mRNA and protein in MCF-7 cells was significantly higher than that in MCF10A cells(both P<0.01).The expression of ZNF460 increased with the decrease of age(P<0.05),histological grade(P<0.01)and TNM stage(P<0.05).After the expression of ZNF460 was down-regulated,the proliferation ability of MCF-7 cells was significantly decreased(P<0.01),and the expression levels of TRIM28 and c-Myc mRNA and protein were significantly decreased(P<0.05).Conclusions ZNF460 is highly expressed in MCF-7 cells,and down-regulation of ZNF460 can significantly inhibit the proliferation of MCF-7 cells.ZNF460 may participate in the malignant progression of breast cancer by promoting the activation of TRIM28-c-Myc axis.
