摘　　要：葡聚糖蔗糖酶是一种糖苷转移酶(Glucosyltransferase,GTF),可以催化蔗糖合成胞外多糖(Exopolysaccharides,EPS)。由于葡聚糖蔗糖酶来源和催化中心的氨基酸序列不同,所以其合成的EPS的结构和性质存在差异。解析GTF的结构和催化机制是探究EPS生物合成和构效关系的关键。本研究利用生物信息学方法对肠膜明串珠菌(Leuconostoc mesenteroides,Ln.mesenteroides)GtfA蛋白序列进行结构和性质分析。结果表明,GtfA属于GH70家族,是一种位于细胞外的亲水蛋白,存在跨膜区,含有19个开放阅读框和7个保守区域,与Ln.mesenteroides CBA3607 Gtf相似性为100%。该蛋白具有一般葡聚糖蔗糖酶的“桶”状结构,其活性口袋能够与蔗糖紧密结合,催化蔗糖合成EPS。本研究结果可为后期探究葡聚糖蔗糖酶的结构和功能提供参考和依据。Glucansucrase is a glycosyltransferase(GTF)which can catalyze sucrose to synthesize exopolysaccharides(EPS).The structure and properties of the synthesized EPS are different due to the the different source and amino acid sequences of catalytic center of glucansucrase.Analysis of the structure and catalytic mechanism of GTF is the key to explore EPS biosynthesis and structure-activity relationship.The structure and properties of Leuconostoc mesenteroides(Ln.mesenteroides)GtfA protein were analyzed by bioinformatic method.The results show that GtfA belongs to GH70 family and is an extracellular hydrophilic protein.The enzyme has transmembrane regions,containing 19 open reading frames and 7 conserved regions.Its similarity to the Ln.mesenteroides CBA3607 Gtf is 100%.The protein has the“barrel-like”structure of common glucansucrase.Its active pockets can bind closely to sucrose and catalyze the sucrose to synthesize EPS.This study results can provide reference and basis for further research on the structure and function of glucansucrase.
