摘 要:本研究以尾叶桉(Eucalyptus urophylla)、毛叶桉(E.torelliana)、窿缘桉(E.exserta)和柠檬桉(E.citriodora)为对象,探究4种桉树叶片挥发性成分的组成与差异,为桉嗜木天牛(Phoracantha semipunctata)的寄主选择提供科学依据。采用聚二甲基硅氧烷/二乙烯苯涂层(PDMS/DVB)、二乙烯苯/Carboxen/聚二甲基硅氧烷涂层(DVB/CAR/PDMS)的萃取和气相色谱-质谱联用法(GC-MS),分析4种桉树叶片挥发性成分。结果表明:2种萃取探头顶空提取4种桉树叶片挥发物的成分具有明显差异,其中尾叶桉叶片挥发物鉴定出56种化合物,主要成分为1,8-桉叶素(相对含量36.27%);毛叶桉叶片挥发物的PDMS/DVB萃取,鉴定出41种化合物,主要成分为左旋-α-蒎烯(相对含量27.73%);窿缘桉叶片挥发物鉴定出52种化合物,主要成分为1,8-桉叶素(相对含量33.44%);柠檬桉叶片挥发物鉴定出36种化合物,主要成分为2,6-二甲基-2,6-辛二烯(相对含量36.24%)。而DVB/CAR/PDMS提取的尾叶桉、毛叶桉、窿缘桉和柠檬桉叶片挥发物分别鉴定出70、58、50和50种化合物,主要挥发性物质分别为1,8-桉叶素(相对含量30.28%)、左旋-α-蒎烯(相对含量29.18%)、(+)-α-蒎烯(相对含量30.69%)和香茅醛(相对含量34.27%)。PDMS/DVB和DVB/CAR/PDMS 2种萃取头检测的挥发性物质主要由烃类和醇类等9类化合物组成。PDMS/DVB和DVB/CAR/PDMS萃取方法在4种桉树叶片挥发性物质组成和主要成分上存在明显差异,DVB/CAR/PDMS萃取方法的检测效果明显优于PDMS/DVB,研究结果对其他树种挥发性物质的检测,以及桉嗜木天牛寄主选择机制的探索具有参考价值。Four Eucalyptus species,including E.urophylla,E.torelliana,E.exserta and E.citriodora were used as the research objects to investigate the composition and differences of volatile components in the leaves so as to provide a scientific basis for the host selection of Phoracantha semipunctata.The volatiles from the leaves were extracted by PDMS/DVB and DVB/CAR/PDMS solid phase microextraction fibers and analyzed by using gas chromatography-mass spectrometry(GC-MS)method.The results indicated that the volatile compounds extracted by the two fibers were different.By using PDMS/DVB fiber,56 components were identified from E.urophylla leaves with the main component of 1,8-cineole,accounting for 36.27%of the total.Comparatively,by using DVB/CAR/PDMS,41 components were identified from E.torelliana leaves,the main component was(-)-α-pinene,accounting for 27.73%of the relative contents of the total identified components;52 components were identified from E.exserta leaves with the main component of 1,8-cineole,accounting for 33.44%of the total;36 components were identified from E.citriodora leaves with the main component of 2,6-dimethyl 2,6-octadiene,accounting for 36.24%of the total;a total of 70,58,50 and 50 compounds were identified from E.torelliana,E.exserta,E.citriodora and E.urophylla,respectively,with the main components of 1,8-cineole(30.28%),(-)-α-pinene(29.18%),(+)-α-pinene(30.69%),citronellal(34.27%)respectively.The volatile compounds extracted by the two fibers were composed of nine classes of compounds,and the main compounds were hydrocarbons and alcohols.The composition and main components of volatile compounds of four Eucalyptus varieties extracted by the two fibers were obviously different,and the detection effect of the DVB/CAR/PDMS was significantly better than that of PDMS/DVB.This is of great value and significance for the detection and analysis of volatile compounds in other tree species and the exploration of the host selection mechanism of P.semipunctata.
