摘 要:穗轴是紫玉米穗脱粒后的剩余部分,其中含有丰富的花色苷,具有广阔的应用前景。本研究采用纤维素酶辅助提取紫玉米穗轴中的花色苷,通过单因素试验确定了料液比、加酶量、提取时间、提取温度4种因素对紫玉米穗轴花色苷提取率的影响规律,并应用正交试验各因素的水平条件进行优化,得到最佳提取工艺:提取时间120 min、提取温度40℃、加酶量20 mg/g、料液比1∶40 g/mL,在此工艺条件下紫玉米穗轴花色苷的提取率达到1.339 mg/g,表明纤维素酶可有效提高紫玉米穗轴花色苷的提取率。本研究进一步运用高分辨率质谱平台LC-MS/MS对获得的紫玉米穗轴花色苷进行分析鉴定,发现含有芍药素3,5-O-二葡萄糖苷等18种花色苷组分。The cob is the remaining part of purple corn after threshing,which is rich in anthocyanins and has broad application prospects.In this study,cellulase was used to assist the extraction of anthocyanins from purple corn cob.The effects of four factors,such as solid-liquid ratio,enzyme addition amount,extraction time and extraction temperature on the extraction rate of anthocyanins from purple corn cob were determined by single factor test.And orthogonal test was applied to optimize the levels of these factors.The optimal extraction conditions were identified as the extraction time of 120 min,the extraction temperature of 40℃,the enzyme dosage of 20 mg/g,and the solid-liquid ratio of 1∶40 g/mL.Under these conditions,the extraction rate of anthocyanins from purple corn cob reached 1.339 mg/g,indicating that cellulase could effectively improve the extraction rate of anthocyanins from purple corn cob.Additionally,the high-resolution liquid chromatography-tandem mass spectrometry(LC-MS/MS)was used to analyze and identify the anthocyanin components in purple corn cob.It was found that it contained 18 anthocyanin components such as peonidin 3,5-O-diglucoside.
