摘 要:为探究威宁黄牛源发酵乳杆菌的潜在益生特性和开发益生菌制剂的可行性,本研究进行了益生菌的分离鉴定、耐酸耐胆盐试验、抑菌试验和全基因组测序研究。结果:从35份样品中分离得到11株发酵乳杆菌,均对低pH值和胆盐有良好的抗性,尤其是发酵乳杆菌5;抑菌试验发现发酵乳杆菌5有显著的抑制大肠杆菌的效果,将其命名为GZHN2023LC。基因组测序发现该菌基因组全长达2077921 bp,共编码2011个蛋白;经京都基因与基因组百科全书(KEGG)、直系同源簇(COG)、基因本体论(GO)、碳水化合物活性酶(CAZy)等数据库比对,发现该基因组碳水化合物和氨基酸代谢能力强,同时转录和膜运输活跃,可能与其发酵多糖产生乳酸以及能够合成抗菌肽、胞外多糖有关。综上,发酵乳杆菌GZHN2023LC能够快速增殖,对酸和耐盐有良好的抗性,且有显著的抗菌特性,基因组测序也发现该菌有良好的益生潜能,具备用于益生菌制剂的潜力。To explore the potential probiotic properties of fermented Lactobacillus isolated from Weining yellow cattle and the feasibility of developing probiotic products,we conducted the isolation and identification of the probiotics,acid and bile salt resistance tests,bacteriostatic tests and whole genome sequencing.The results showed that 11 strains of Lactobacillus fermentum were isolated from 35 samples,and the strains had good resistance to low pH and bile salts,especially Lactobacillus fermentum 5.The bacteriostatic test showed that Lactobacillus fermentum 5 had significant inhibition effect on Escherichia coli,which was named GZHN2023LC.Genome sequencing revealed that the total length of the bacterial genome was 2077921 bp,encoding a total of 2011 proteins.Through comparison with databases such as Kyoto Ency⁃clopedia of Genes and Genomes(KEGG),Direct Homologous Clusters(COG),Gene Ontology(GO),Carbohydrate Active Enzymes(CAZy)and other databases,it was found that this genome had strong metabolic capacity of carbohydrates and amino acids,and active tran⁃scription and membrane transport,which might be related to the production of lactic acid by fermentation polysaccharide and the synthesis of antimicrobial peptides and extracellular polysaccharides.This study confirmed that Lactobacillus fermentum GZHN2023LC multiplied rapidly,had good resistance to acid and salt,and had significant antibacterial properties.Genome sequencing also found that this bacterium had good probiotic potential,and could be used in probiotic products.
