摘  要:以白木香渣为原料,采用超微破壁及复合酶(纤维素酶、半纤维素酶)技术探索沉香油提取新工艺。在单因素实验基础上选取白木香渣粒度、提取时间、纤维素酶比例为影响因子,采用Box-Behnken试验设计,以沉香油得率为响应值进行分析,并应用GC-MS进行成分检测。最优工艺为:酶添加量50 U/g、酶解时间3 h、底物浓度20%、纤维素酶比例56%、提取时间3.15 h、粉碎粒度300~400目,此工艺条件下得率为12.10%;传统方法提取的沉香油成分主要含色酮(33.16%)和香豆素(22.23%);优化方法提取的沉香油成分主要含色酮(23.92%)和香豆素(24.49%),优化提取的沉香油中组分总含量高出传统提取方法组分含量57.25%,且沉香油色泽显著优于传统方法(P<0.05)。优化的提取工艺合理、可行,能有效提高沉香渣中沉香油得率。A new extraction process of agarwood oil was explored by using Aquilaria sinensis(Lour.)Gilg residue as the raw material,combined with ultra-micro-broken walls and compound enzymes(cellulase and hemicellulose).Based on the single factor experiments,select the particle size of agarwood residue,extraction time and cellulase ratio as the influence factor,the yield of essential oil were examined by the response surface method,the multiple regression model was established for response analysis,colorimetric detection and GC-MS detection.The optimum process was:enzyme addition 50 U/g,enzyme hydrolysis time 3 h,substrate concentration 20%,cellulase ratio 56%,extraction time 3.15 h,grinding granularity 300-400 mesh,under this process parameter the extraction rate was 12.10%.Agarwood oils mainly contains chromone(33.16%)and coumarin(22.23%)extraction by the traditional method,while by the optimized method mainly contains chromone(23.92%)and coumarin(24.49%).The total component content of the optimized agarwood oil was 57.25%higher than that of the traditional method,and the color of agarwood oil was significantly better than that of the traditional method(P<0.05).The extraction process of essential oil from agarwood is reasonable and feasible,it can effectively improve the extraction rate of agarwood oil.
