摘 要:黄姑鱼是我国重要的海水经济鱼类,随着养殖规模逐渐扩大,对其优良养殖种质的需求也愈加迫切。不饱和脂肪酸含量作为重要的营养品质性状,受到育种研究学者的广泛关注。测定了179尾黄姑鱼选育群体后代的肌肉脂肪酸组成,获得了以n-3长链多不饱和脂肪酸相对含量等表型性状,对这179尾个体开展全基因组重测序,经过质控、变异检测后获得2 940 251个高质量单核苷酸多态性(SNP)位点;基于混合线性模型对脂肪酸表型开展全基因组关联分析,定位到19个显著相关的SNP位点,并鉴别到27个候选基因,其中编码Mth938结构域包含蛋白的基因(aamdc)被认为是参与调控脂肪生成过程,可能是后续研究黄姑鱼肌肉脂肪酸组成以及进行相关选育研究的重要标记基因。研究结果为解析鱼类肌肉脂肪酸组成的遗传机制研究提供了参考依据,同时也为黄姑鱼优良新品种培育提供了理论指导。Yellow drum(Nibea albiflora)is an important marine economic fish species in China.As the scale of breeding gradually expands,the demand for new high-quality breeding germplasm becomes urgent.Unsaturated fatty acid content,as an important nutritional quality trait,is also gaining widespread attention from the researchers.The offspring of a breeding population consisting of 179 yellow drums was analyzed by whole-genome re-sequencing.As a result,2940251 high-quality single nucleotide polymorphism(SNP)loci were obtained.The fatty acid composition of each sample was determined,and with the traits such as n-3 long-chain polyunsaturated fatty acid content as phenotypic characteristics,a genome-wide association analysis was performed based on a mixed linear model,and the molecular markers related to the muscle fatty acid composition in yellow drum were identified.A total of 19 significantly-associated SNP loci were pinpointed,and 27 candidate genes were screened,in which the aamdc(Mth938 domain-containing protein)gene was considered in previous studies as being involved in regulating the fat synthesis process,which may serve as an important marker gene for subsequent research on the muscle fatty acid composition and related breeding studies in yellow drum.This study provided a reference for the genetic mechanism research of fish muscle fatty acid composition and also a theoretical guidance for the breeding of superior new varieties of yellow drum.
