摘 要:卵巢是影响蛋鸡产蛋性能的重要器官,产蛋期卵巢生理机能衰退会导致产蛋率下降。本文旨在研究不同水平虾青素对13月龄罗曼粉蛋鸡卵巢组织和Sirtuin基因家族表达的影响。选取16只健康、体重相近的13月龄罗曼粉蛋鸡,随机分为4组,每组4个重复,分别为对照B组、试验LC组、MC组和HC组。所有蛋鸡饲喂相同的基础日粮,每日饲喂三次。对照组B额外灌喂葵花籽油4mL,试验组LC、MC和HC分别额外灌喂4、20、100mg/kg体重的虾青素(溶剂为葵花籽油)4mL。饲喂方式为每天下午18:00直接灌服。各组温度保持在(25±3)℃,湿度维持在65%~80%,试验期25 d。结果显示,灌喂虾青素会使蛋鸡卵巢组织结构着色点增多、着色加深,卵泡壁增厚,卵泡数量有所增加。试验MC组卵巢组织中的Sirt2、Sirt3基因表达量极显著高于对照组(P<0.01),Sirt6、Sirt7基因表达量显著高于对照组(P<0.05),LC组卵巢组织中的Sirt1基因表达量极显著高于对照组(P<0.01)。综上所述,虾青素能够改善蛋鸡卵巢组织结构,延缓卵泡衰老,促进卵巢中Sirtuin基因家族的表达,其中20mg/kg的剂量效果最佳。本试验结果可为虾青素调控卵巢生理功能机制的研究提供一定的参考。The ovary is an important organ which affects laying performance of laying hens.The decline of physiological function of ovary during laying period will lead to the decrease of laying rate.The aim of this study was to investigate the effects of different levels of astaxanthin on ovarian tissue structure and Sirtuin gene expression in 13-month-old Roman laying hens.16 healthy 13-month-old Roman laying hens with similar body weight were randomly divided into 4 groups with 4 replicates in each group,The 4 groups were control group B,experimental group LC,MC and HC respectively.All layers were fed the same basic diet three times a day.The control group was fed with 4 mL sunflower seed oil,and the experimental groups(LC,MC,HC)were fed with 4 mL of astaxanthin-sunflower oil mixture at 4,20 and 100 mg/kg body weight,respectively.Hens were perfused with astaxanthin at 18:00 pm every day.The temperature and humidity of each group were maintained at(25±3)℃and 65%~80%respectively for 25 days.The results showed that administration of astaxanthin could increase the staining spots,deepen the staining spots,thicken the wall of follicles and increase the number of follicles.The expression levels of Sirt2 and Sirt3 in MC were significantly higher than those in control group(P<0.01).The expression levels of Sirt6 and Sirt7 in MC ovarian tissues were significantly higher than those in control group(P<0.05).The expression level of Sirt1 gene in LC ovarian tissues was significantly higher than that in control group(P<0.01).In conclusion,Astaxanthin can improve ovarian tissue structure,delay follicle senility and promote the expression of Sirtuin gene family in ovary of laying hens,and the dose of 20 mg/kg has the best effect.These results provide reference for studying the regulatory mechanism of astaxanthin on ovary function of hens.
