摘  要:以地骨皮为研究对象,优化间歇式超声辅助酶法提取地骨皮多糖工艺条件,并初步评价其体外抗氧化能力。以单因素试验结果为基础,采用响应面法进一步考察酶添加量、酶解pH、超声功率、超声温度对地骨皮多糖得率的影响,并通过地骨皮多糖对1,1-二苯基-2-三硝基苯肼(DPPH)自由基、羟基自由基和超氧阴离子自由基的清除能力,评价其抗氧化能力。结果表明,在酶添加量2.5%,酶解pH4.6,超声功率167W,超声温度50℃,超声累计时间30min的最优提取工艺条件下,地骨皮多糖得率为18.63%,纯度为87.32%;抗氧化试验结果表明,地骨皮多糖对DPPH自由基、羟基自由基和超氧阴离子自由基的清除能力均较好。综上可知,采用间歇式超声辅助酶法提取地骨皮多糖工艺可行、可控,所得多糖抗氧化能力良好。In this study,the extraction conditions of polysaccharides from Cortex lyci were optimized by intermittent ultrasound-assisted enzymatic method,and the antioxidant capacities in vitro were evaluated.On the basis of single factor tests,the response surface method was used to further investigate the effects of enzyme additions,enzymatic pH values,ultrasonic power,and ultrasonic temperatures on the yields of crude polysaccharides,then the antioxidant activities of crude polysaccharides were evaluated by the scavenging abilities of DPPH,hydroxyl and superoxide anion radicals.The results showed that under the following optimal extraction conditions(enzyme addition of 2.5%,enzymatic pH value of 4.6,ultrasonic power of 167 W,ultrasonic temperature of 50℃C,and ultrasonic cumulative time of 30 min),the yield of polysaccharides was 18.63%with purity of 87.32%.The results of antioxidant test showed that the Cortex lyci crude polysaccharides presented superior antioxidant capacities to scavenging DPPH,hydroxyl and superoxide anion radicals.In conclusion,the intermittent ultrasound-assisted enzymatic extraction of Cortex lyci polysaccharides is feasible and controllable,and the obtained polysaccharides have good antioxidant capacities.
