摘  要:目的:建立一种利用高效液相色谱技术检测食用油内黄曲霉毒素B_(1)含量的方法。方法:以甲醇-水体系对样品进行萃取,通过黄曲霉毒素B_(1)特异性免疫亲和柱进行净化处理,并以甲醇作为洗脱剂进行洗脱,使用高效液相色谱仪进行检测分析。结果:黄曲霉毒素B_(1)浓度在0.1~40.0 ng·mL^(-1)线性关系良好,相关系数为0.999 6;方法的平均回收率在97.2%~100.3%,相对标准偏差在0.4%~0.9%;方法检出限为0.029 μg·kg^(-1),定量限为0.098 μg·kg^(-1),均满足检测要求。结论:该方法快速高效、操作简便,且试剂耗材消耗少,适用于检测食用油中黄曲霉毒素B_(1)的含量。Objective:To establish a method for the determination of aflatoxin B_(1) in edible oil by high performance liquid chromatography (HPLC).Method:The samples were extracted by methanol-water system,purified by aflatoxin B_(1) specific immunoaffinity column,eluted with methanol as eluent,and analyzed by HPLC.Result:Aflatoxin B_(1) concentration has a good linear relationship in the range of 0.1 ng·mL~(-1) to 40.0 ng·mL~(-1),the correlation coefficient was 0.999 6;the average recoveries ranged from 97.2% to 100.3%,and the relative standard deviations ranged from 0.4% to 0.9%;the limits of detection and quantitation were 0.029 μg·kg~(-1) and 0.098 μg·kg~(-1),both of which met the detection requirements.Conclusion:The method is fast,efficient,easy to operate,with less consumption of reagents and consumables.It is suitable for the determination of aflatoxin B_(1) in edible oil.
