益生菌群半固态发酵对甘草中有效成分释放及其活性的影响

摘  要:甘草植物细胞壁阻碍了胞内甘草酸、黄酮等有效成分的释出,本文利用酿酒酵母菌、乳酸菌、芽孢杆菌组成的益生菌群半固态发酵作用,增加胞内活性物质的释放,并对发酵液的抑菌、抗氧化能力和美白潜力进行了评价。结果表明,益生菌群发酵后甘草多糖、黄酮、甘草酸和甘草次酸的提取率与索氏提取法相当,发酵液对大肠杆菌、肺炎克雷伯杆菌、鲍曼不动杆菌的抑菌率分别为73.85%、45.57%、39.92%,对1,1-二苯基-2-三硝基苯肼(DPPH)、2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)、2-苯基-4,4,5,5-四甲基咪唑啉-3-氧代-1-氧(PTIO)、超氧基自由基、羟基自由基的清除效果优于其他方法,如清除PTIO的能力分别是索氏提取、纯水摇瓶提取和三种菌单独发酵的1.71、2.69、2.00、1.00和2.11倍,对酪氨酸酶抑制率为61.62%。甘草的益生菌群发酵液为保健品和化妆品的开发提供了优质的原料。The cell wall of licorice prevents the release of effective substances such as flavonoids and polysaccharides from liquorice cells.In this study,a mixed probiotics consortium composed of Saccharomyces cerevisiae,lactic acid bacteria and Bacillus was used in semisolid-sate fermentation of liquorice powder to promote the release of effective substances.The inhibition ability on pathogenic bacteria and tyrosinase as well as antioxidant activities of the fermentation broth were evaluated.The experimental results showed that the extraction rates of polysaccharides,flavonoids,glycyrrhizic acid and glycyrrhetinic acid could be considerably compared with the Soxhlet method.The inhibition rate of the mixed fermentation supernatant on Escherichia coli,Klebsiella pneumoniae and Acinetobacter baumannii was 73.85%,45.57%and 39.92%,respectively.The scavenging rates of the mixed fermentation supernatant to 1,1-diphenyl-2-picrylhydrazyl(DPPH),2,2′-Azinobis-(3-ethylbenzthiazoline-6-sulphonate(ABTS),2-Phenyl-4,4,5,5-tetramethylimidazoline-3-oxide-1-oxyl(PTIO),superoxide radical and hydroxyl radical were superior to other extraction methods.The inhibition rate on tyrosinase was 61.62%.The semisolid-state fermentation of licorice by probiotic consortium provides high-quality raw materials for the development of health-care products and cosmetics.
