摘 要:糖尿病视网膜病变(DR)是糖尿病患者常见的眼部并发症,也是导致成人失明的主要原因之一。由于DR的复杂病理机制及其与其他代谢紊乱的紧密联系,探索其病因并开发有效的预防和治疗手段成为公共卫生领域的紧迫需求。孟德尔随机化作为一种新兴的因果推断方法,通过利用遗传变异来推断暴露与疾病之间的因果关系,能够有效控制混杂因素和逆因果关系的影响。本述评旨在介绍孟德尔随机化及其在DR病因探索中的应用和研究进展,同时分析当前研究的局限性与未来发展方向。Diabetic retinopathy(DR)is a common ocular complication in patients with diabetes and one of the leading causes of blindness in adults.Due to the complex pathological mechanism of DR and its close relationship with other metabolic disorders,it has become an urgent need in the field of public health to explore its etiology and develop effective prevention and treatment methods.As a new causal inference method,Mendelian randomization(MR)can effectively control the influence of confounding factors and inverse causality by using genetic variations to infer the causal relationship between exposure and diseases.The purpose of this paper is to introduce the application and research progress of MR in the investigation of the etiology of DR and analyze the limitations of current research and the future development direction.
