摘  要:目的 观察膜活化剂吐温 80合并温热作用对人胃癌细胞BGC 82 3的抑制效应及其对Hsp70、bcl 2、bax表达的影响。方法 通过MTT法 ,测定 0 .1%及 0 .15 %吐温 80与 4 2℃温热合并作用对BGC 82 3细胞的抑制作用 ;运用免疫细胞化学染色 ,观察 0 .1%吐温 80与 4 2℃温热合并作用后 (0h、8h、16h、2 4h)BGC 82 3细胞Hsp70、bcl 2、bax表达的改变。结果 吐温 80合并 4 2℃10 0min温热作用对BGC 82 3细胞具有明显的抑制效应 ,温热后保留吐温 80继续作用将进一步增强抑制效应 ,合并抑制效应与吐温 80浓度相关 (P <0 .0 1)。合并作用能明显抑制BGC 82 3细胞热作用后的Hsp70表达 ,但bcl 2、bax的表达均增加 ,且bax的表达强于bcl 2 ,并全部分布于胞浆。结论 吐温 80与 4 2℃温热有明显协同抑制肿瘤细胞生长效应 ,对Hsp70和bcl 2、bax表达及分布的影响提示其抑制机理可能与诱导肿瘤细胞凋亡有关。Objective To investigate the effects of hyperthermia at 42℃ for 100 min with Tween 80 on the cell viability and expression of apoptosis related genes in human gastric cancer cell line BGC 823. Methods Cell viability was examined by methyl tetrazolium (MTT) assay. The expression of hsp70, bcl 2 and bax was determined by the immunocytochemical method at 0h, 8h, 16h and 32h after treatment respectively. Results Hyperthermia at 42℃ for 100 min with Tween 80 markedly inhibited the cell growth. The inhibitory effect was obviously intensified by maintaining Tween 80 for 24h after heating. The protein levels of bcl 2 and bax were elevated while expression of hsp70 decreased significantly. The expression of bax was stronger than that of bcl 2, and distributed in the cytoplasm. Conclusion Hyperthermia at 42℃ for 100 min combined with Tween 80 can suppress growth of gastric cancer cells through induction of apoptosis which may be involved in different expressions of apoptosis related genes.
