溃疡性结肠炎的IL-1β、IL-1RA、L MP2基因多态性研究

摘　　要：目的 :研究溃疡性结肠炎 (UC)病人的IL 1β、IL 1RA、LMP2基因多态性 ,并分析其与抗中性粒细胞胞浆抗体 (AN CA)及临床分型的关系。方法 :用PCR 限制性片段多态性 (PCR RFLP)方法和序列特异性引物 PCR(PCR SSP)方法分别对 81例UC病人和 114名健康者进行IL 1β、LMP2和IL 1RA基因多态性分析。结果 :UC病人和健康者之间IL 1β、IL 1RA和LMP2各基因型和基因频率比较均无显著性差异 (P >0 0 5 ) ;当UC病人分为ANCA(+)组和ANCA(- )组后 ,发现ANCA(+)组IL 1RA等位基因 2频率高于ANCA(- )组 (13 7%对 3 3 % ,P <0 0 5 ) ,其它各基因型及基因频率比较 ,统计学上均无显著性差异 (P>0 0 5 )。结论 :中国汉族UC病人与IL 1β、IL 1RA、LMP2基因多态性无关联 ,但ANCA(+)UC病人IL 1RA等位基因 2频率明显增加。Objective:To know whether the Chinese UC patients are associated with IL-1β, IL-1RA, LMP2 gene polymorphisms and to analyze the relationship among gene polymorphisms, antineutrophil cytoplasmic antibodies (ANCA) and clinic classification of ulcerative colitis (UC).Methods:81 UC patients, 114 healthy subjects (HS) were enrolled in the study,IL-1β, LMP2 and IL-1RA gene polymorphisms were analyzed with polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and polymerase chain reaction with sequence-specific primers (PCR-SSP) respectively.Results:There were not difference in the genotype and the gene frequencies of IL-1β, IL-1RA and LMP2 between UC patients and HS (P>0.05). When UC patients were divided into ANCA positive and negative groups, it was found that the frequency of allele 2 of IL-1RA in ANCA positive UC patients was significantly higher than that in ANCA negative UC patients (13.7% versus 3.3%, P<0.05), there were not significant difference on the other genotype and gene frequencies of allele (P>0.05).Conclusion:The Chinese UC patients are not associated with IL-1β, IL-1RA and LMP2 gene polymorphisms, but the frequency of allele 2 of IL-1RA in ANCA positive UC patients was significantly increased.
