响应面法优化毛木耳粗多糖提取工艺及其抗肿瘤活性评价

摘  要:采用热水浸提法提取毛木耳粗多糖。以粗多糖得率、总糖含量、生物活性评价为权重指标计算综合评分,采用单因素和响应面试验优化毛木耳粗多糖提取工艺,并评价其抗肿瘤活性。结果表明:最优提取工艺参数为粉碎粒径0.125mm、提取溶液pH9、提取1次、提取温度60℃、料液比1:100(g/mL)、提取时间60min,在此条件下,毛木耳粗多糖的综合评分为64%。抗肿瘤活性试验结果显示,毛木耳粗多糖明显降低了4T1乳腺癌小鼠的肿瘤质量和肺转移结节数,表明其在抗肿瘤活性方面有较好的作用。The crude polysaccharide from Auricularia polytricha was extracted by hot water extraction.The comprehensive score was calculated based on the yield of crude polysaccharide,total sugar content,and biological activity evaluation as weighting indicators.Single factor experiments and response surface experiment were used to optimize the extraction process of the crude polysaccharide from Auricularia polytricha and its antitumor activity was evaluated.The results showed that the optimal extraction parame-ters were crushed particle size 0.125 mm,extraaction solution pH 9,extraction once,extraction temper-ature 60℃,solid-to-liquid ratio 1:100(g/mL),and extraction time60min.Under theseconditions,the comprehensive score of the crude polysaccharide from Auricularia polytricha was 64%.The antitu-mor activity assay result demonstrated that the crude polysaccharide from Auricularia polytricha signifi-cantly reduced the tumor mass and the number of lung metastatic nodules in 4T1 breast cancer mice,in-dicating it had a good role in antitumor activity.
