摘 要:为探究臭常山(Orixa japonica)提取物的体外抑菌效果及其活性成分,以臭常山为试验材料,采用60%乙醇溶液、石油醚、乙酸乙酯和丙酮获得提取物和水层物质,并制备臭常山水提取物,采用纸片扩散法(K-B法)评估臭常山不同溶剂提取物的抑菌活性;采用质谱法(MS)鉴定活性成分。结果表明,臭常山不同溶剂提取物对大肠杆菌(Escherichia coli)、铜绿假单胞菌(Pseudomonas aeruginosa)、粪肠球菌(Enterococcus faecalis)、普通变形杆菌(Proteus vulgaris)和肺炎克雷伯氏菌(Klebsiella pneumoniae)的抑制作用均不明显;60%乙醇提取物、乙酸乙酯提取物和丙酮提取物对金黄色葡萄球菌(Staphylococcus aureus)的抑制作用均较好;60%乙醇提取物对金黄色葡萄球菌的抑制作用最好,抑菌圈直径为14.00 mm;乙酸乙酯提取物的最小抑菌浓度(MIC)为0.25 g/mL。60%乙醇提取物对燕麦镰刀菌(Fusarium avenaceum)、尖孢镰刀菌(Fusarium oxysporum)和层出镰刀菌(Fusarium proliferatum)均有一定抑制作用,平均抑菌率为16.67%。在乙酸乙酯提取物中鉴定出14种化合物,其抑菌作用可能与其中含有多种生物碱有关。To investigate in vitro antibacterial effects of Orixa japonica extracts and their active components,O.japonica was used as experimental material.Extracts and water-soluble substance were obtained by using 60%ethanol solution,petroleum ether,ethyl acetate and acetone.Water extract of O.japonica was prepared.Antibacterial activity of extracts from different solvents was evaluated by disk diffusion(K-B method).Active components were identified by mass spectrometry(MS).Results showed that O.japonica extracts from different solvents did not exhibit obivious inhibitory effects against Escherichia coli,Pseudomonas aeruginosa,Enterococcus faecalis,Proteus vulgaris and Klebsiella pneumoniae.60%ethanol extract,ethyl acetate extract and acetone extract had relative good inhibitory effects against Staphylococcus aureus.60%ethanol extract exhibited the best inhibitory effect against S.aureus with inhibition zone diameter of 14.00 mm.Minimum inhibitory concentration(MIC)of ethyl acetate extract was 0.25 g/mL.60%ethanol extract showed certain inhibitory effects against Fusarium avenaceum,Fusarium oxysporum,and Fusarium proliferatum with average inhibition rate of 16.67%.14 compounds were identified in ethyl acetate extract,and antibacterial effects of ethyl acetate extact may be related to various alkaloids in extract.
