摘 要:为揭示木棉(Bombax ceiba)花颜色差异的代谢机理,为后续木棉新品种创制研究提供理论依据,以深红色、红色、橙红色、橙黄色和黄色木棉花花瓣为试验材料,采用高效液相色谱-串联质谱法(HPLC-MS/MS),对5种木棉花花瓣进行代谢组学测定,分析花瓣黄酮类物质组成和含量。结果表明,共鉴定出6类137种黄酮类代谢物,分别为黄酮和黄酮醇(79种)、黄烷酮(19种)、花青素(14种)、异黄酮(12种)、查耳酮和二氢查耳酮(7种)及类黄酮(6种)。黄酮和黄酮醇代谢物在5种颜色花瓣中含量均最高,占比均超过65%;其次为黄烷酮和花青素代谢物。花青素代谢物分析显示,花青素苷、矢车菊素3-O-芸香糖苷、矢车菊素-3-半乳糖苷和矢车菊素3-O-葡萄糖苷在木棉深红色花瓣中含量均较高。KEGG分析显示,5种颜色花瓣的差异代谢物均显著富集在花青素合成通路上。影响木棉花花瓣呈红色的主要代谢物质是花青素,呈现黄色可能为高含量的黄酮和黄酮醇代谢物及低含量的花青素类物质。In order to reveal metabolic mechanisms underlying flower color differences in Bombax ceiba and provide theoretical basis for development of new cultivars,B.ceiba petals with five different colors including dark red,red,orange red,orange yellow and yellow were used as experimental materials.Metabolomic analysis was performed on B.ceiba petals using high performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS)technology to analyze compositions and contents of flavonoids.Results showed that total of 137 flavonoids metabolites from six classes were identified,including flavones and flavonols(79),flavanones(19),anthocyanins(14),isoflavonoids(12),chalcones and dihydrochalcones(7)and flavonoids(6).Contents of flavonoids and flavonols metabolites were the highest in all five petals,accounting for more than 65%,followed by flavanones and anthocyanins metabolites.Anthocyanin metabolie analysis revealed that cyanin chloride,keracyanin chloride,idaein chloride and cyanidin 3-O-glucoside were the highest in dark red petals of B.ceiba.KEGG analysis indicated that differential metabolites in all five petals were significantly enriched in anthocyanin synthesis pathway of anthocyanin.Primary metabolites responsible for red coloration of B.ceiba petals were anthocyanins,while yellow coloration might be associated with high levels of flavones and flavonols and low levels of anthocyanins.
