摘  要:目的:优化超声辅助乙醇溶剂法提取牡丹花残渣提取物中总黄酮和总多酚的工艺条件,并对其抗氧化活性进行研究。方法:以牡丹花残渣提取物中总黄酮和总多酚得率为评价指标,采用单因素和响应面法优化其提取工艺,并通过DPPH法、ABTS法和Fe3+还原能力法评价其抗氧化活性。结果:牡丹花残渣提取物中总黄酮和总多酚提取最佳工艺条件为乙醇浓度70%、液料比42mL/g、超声温度41℃。此条件下牡丹花残渣提取物中总黄酮和总多酚提取率分别为0.72%和2.51%。抗氧化活性实验表明,牡丹花残渣提取物在1.0mg/mL条件下对DPPH、ABTS自由基清除能力和Fe^(3+)还原力吸光度值为73.96%、100.00%、1.204。结论:牡丹花残渣提取物中活性成分具有显著抗氧化活性,可为牡丹花残渣的开发利用提供理论参考。Objective:To optimize the process conditions for extracting total flavonoids and total polyphenols from residue extract of the flowers of Paeonia suffruticosa by ultrasound-assisted ethanol solvent method and study their antioxidant activities.Methods:With the extraction yields of total flavonoids and total polyphenols in the residue extract from the flowers of P.suffruticosa as the evaluation indexes,single-factor experiments and response surface methodology were used to optimize the extraction process.The antioxidant activity was evaluated by DPPH radical scavenging activity,ABTS radical scavenging activity and Fe^(3+) reducing activity.Results:The optimal process conditions for the extraction of total flavonoids and total polyphenols from residue extract of the flowers of P.suffruticosa were ethanol concentration of 70%,liquid-solid ratio of 42 mL/g,and extraction temperature of 41℃.Under these conditions,the extraction yields of total flavonoids and total polyphenols were 0.72% and 2.51%,respectively.The results of antioxidant activity experiments showed that at a concentration of 1.0 mg/mL,the DPPH radical scavenging ability,ABTS radical scavenging ability and Fe3+reducing power of the residue extract from the flowers of P.suffruticosa were 73.96%,100.00%,and 1.204,respectively.Conclusion:Residue extract from the flowers of P.suffruticosa had high content of active ingredients and antioxidant activity.It could provide theoretical reference for the development and utilization of residue from the flowers of P.suffruticosa.
