摘 要:阿尔茨海默病(AD)是临床最常见的神经退行性疾病,其发病受环境和遗传因素的共同影响。2型类髓系细胞触发受体基因(TREML2)是新发现的TREM家族重要成员之一,其与AD的关联逐渐得到了学界的关注。TREML2基因变异与AD易感风险及AD相关内表型密切相关;其编码的免疫受体TREML2可调控小胶质细胞活化状态及功能参与AD病理进程。文中从遗传、临床及分子生物学的角度,结合本课题组前期研究成果,对TREML2在AD中的研究成果进行了综述,期待能为后续AD的遗传及发病机制研究及治疗靶点开发提供借鉴与参考。Alzheimer’s disease(AD)is the most common neurodegenerative disease,and its pathogenesis is influenced by both environmental and genetic factors.The triggering receptor expressed on myeloid cells like transcript 2 gene(TREML2)is one of the newly identified important members of the TREM family,and its association with AD has been paid more attention in recent years.The genetic variation of TREML2 is closely related to AD susceptibility and AD-related endophenotypes.TREML2 encodes an immune receptor that can regulate the activation status and function of microglia,participating in the pathological process of AD.The recent findings of TREML2 in AD from the perspectives of genetics,clinical,and molecular biology,combined with the previous observations from our research group were comprehensively summarized to provide more references for the upcoming research on the genetics and pathogenesis of AD and the development of novel therapeutic targets.
