摘  要:为了建立一种准确、快速、高效的呕吐毒素免疫荧光传感器(DON-icELISA),试验以呕吐毒素(deoxynivalenol,DON)为研究对象,利用二氧化锰包金纳米片(AuNP@MnO_(2))独特的光学特性作为荧光猝灭剂。结果表明,试验建立的DON-icELISA方法线性回归方程为Y=54.116 X+110.53(R 2=0.9906)。检测范围(IC_(20)~IC_(80))为0.021~0.273 ng/mL,检测限为0.013 ng/mL,且方法准确度较高。将所建立的DON-icELISA用于DON及其结构类似物3-脱氧雪腐镰刀菌烯醇(3-DON)、5-乙酰脱氧雪腐镰刀菌烯醇(5-DON)、黄曲霉毒素B1(aflatoxin B1,AFB1)、伏马菌素B1(fumonisin B1,FB1)、玉米赤霉烯酮(zearalenone,ZEN)、T-2毒素和赭曲霉毒素A(ochratoxin A,OTA)均无交叉反应。采用市售玉米面作为待检测食品,采用高(270 pg/mL)、中(150 pg/mL)、低(30 pg/mL)剂量进行加标回收测试该方法的回收率为94.74%~103.46%,平均变异系数为5.88%,通过3名不同专业试验人员进行加标回收试验,变异系数均小于5.00%,说明建立的DON-icELISA具有特异性好、敏感性高、操作简单且结果稳定的优势,可用于DON的快速筛检。This test focused on deoxynivalenol(DON)as the research object and utilized the unique optical properties of MnO 2 nanosheets that wrapped around gold particles(AuNP@MnO_(2))as the fluorescence quencher.This study established an accurate,fast,and efficient DON immunofluorescence sensor(DON-icELISA).Results showed that the linear regression equation of the method established in this test was Y=54.116 X+110.53(R 2=0.9906).The detection range(IC_(20)-IC_(80))was 0.021-0.273 ng/mL,and the detection limit was 0.013 ng/mL.The established DON-icELISA was applied to tests of DON and its structural analogues 3-DON,5-DON,AFB1,FB1,ZEN,T-2,and OTA,without cross reactivity.Using commercially available corn flour as the food sample,the high(270 pg/mL),medium(150 pg/mL),and low(30 pg/mL)doses were used for the recovery test.The recovery rate of this method ranged from 94.74%to 103.46%,the average coefficient of variation was 5.88%.The coefficient of variation was less than 5.00%for the spiked recovery test conducted by three different professional testers.It indicated that DON-icELISA established in this test showed advantages of good specificity,high sensitivity,simple operation with stable results,and could be used for rapid screening of DON.
