摘 要:目的:研究中药材千里光的叶绿体基因组特征,为千里光属植物的系统发育关系和进化研究提供依据。方法:通过高通量测序获得千里光的叶绿体基因组,Codon W、MISA、REPuter、MAFFT等软件分析其密码子偏好、SSR、重复序列、核苷酸多态性、系统发育等特征。结果:千里光叶绿体基因组大小为150 456 bp,编码133个基因,其中蛋白质编码基因88个,偏好使用A/U结尾的密码子。千里光属植物叶绿体基因组的简单重复序列以A/T型单核苷酸重复最多,散在重复序列主要类型为正向重复和回文重复。IR边界和基因组序列比对分析显示千里光属植物有一定的保守性,SSC区的变异程度最高。系统发育分析结果显示千里光属植物分为2个分支,千里光与Senecio asperifolius和Senecio nudicaulis的亲缘关系较近。结论:对千里光属植物叶绿体基因组特征及系统发育的比较研究,为千里光属植物的分子鉴定、遗传多样性和系统进化研究提供了依据。Objective:The chloroplast(cp)genome sequence of the Chinese medicinal material Senecio scandens was determined.The cp genome characteristics and phylogenetic relationship of Senecio plants were investigated.Methods:The cp genome of S.scandens was obtained by high throughput sequencing.The characteristics of codon bias,SSR,repeat sequences,nucleotide polymorphism,and phylogeny were analyzed by Codon W,MISA,REPuter,MAFFT,RAxML-NG and MrBayes software,respectively.Results:The cp genome of S.scandens was 150456 bp in size,encoding a total of 133 genes,including 88 protein coding genes,which prefer to use codons ending in A/U.The SSRs in cp genomes of Senecio plants were predominantly A/T type single nucleotide repeats.The primary types of repetitive sequences were forward repeats and palindrome repeats.The analysis of IR boundary and genome sequence alignments indicated that the Senecio plants were somewhat conservative,with SSC region exhibiting the highest degree of variation.The phylogenetic analysis showed that Senecio plants could be divided into 2 clades,and S.scandens was closely related to Senecio asperifolius and Senecio nudicaulis.Conclusion:The comparative analysis on the cp genome characteristics and phylogeny of Senecio plants would provide a basis for further study on their molecular identification,genetic diversity,and phylogeny.
