摘  要:目的:探讨新型明胶/聚己内酯(GE/PCL)纳米纤维支架(简称GE/PCL支架)复合重组人骨形态发生蛋白-2(rhBMP-2)促骨组织再生作用的机制。方法:从ATCC细胞库中购买原代人源性成骨细胞,传至第三代,通过MTT法确定最佳铺板浓度,在96孔板中培养,与rhBMP-2/GE/PCL复合支架等材料共同培养,然后观察细胞增殖、分化,用CCK8法检测细胞增殖情况,用Western blot技术从蛋白质水平分析不同材料对成骨细胞中相关蛋白表达的影响。结果:rhBMP-2释放量随着时间的延长而增加。rhBMP-2/GE/PCL复合支架和rhBMP-2/Bio-Gide复合膜在促进成骨细胞增殖上效果显著,与对照组相比具有统计学差异。而GE/PCL支架和Bio-Gide膜效果稍弱。此外,rhBMP-2/Bio-Gide复合膜与rhBMP-2/GE/PCL复合支架组在各时间点可显著促进成骨相关蛋白Sp7和Runx-2的表达,与GE/PCL相比均具有显著性差异;但这两种复合材料之间差异不显著。结论:rhBMP-2/GE/PCL复合支架与rhBMP-2/Bio-Gide复合膜可促进成骨细胞增殖与成骨蛋白的表达,具备显著的骨组织再生潜力。Objective:To investigate the mechanism of new gelatin/polycaprolactone(GE/PCL)nanofibrous scaffold combined with recombinant human bone morphogenetic protein-2(rhBMP-2)in promoting bone regeneration.Methods:The primary human osteoblasts were purchased from ATCC cell library and passaged to the third generation.The optimal plate concentration was determined by MTT method. The osteoblasts cells were cultured in 96-well plates and co-cultured with rhBMP-2/GE/PCL scaffolds and other materials. Afterwards, cell proliferation and differentiation were detected by CCK 8 method, and western blot was used to analyze the effects of different materials on the expressions of related proteins in osteoblasts at the protein level.Results: The release of rhBMP-2 increased with the time. The rhBMP-2/GE/PCL composite scaffold and rhBMP-2/Bio-Gide composite membrane had a significant effect on promoting the proliferation of osteoblasts, which was statistically significant compared with the control group. However, GE/PCL scaffold and Bio-Gide membrane were slightly less effective. In addition, rhBMP-2/Bio-Gide composite membrane and rhBMP-2/GE/PCL composite scaffold could significantly promote the expressions of osteogenesis-related proteins Sp7 and Runx-2 at each time point, which were significantly different from those of GE/PCL. However, there was no significant difference between the two composites.Conclusion: rhBMP-2/GE/PCL composite scaffold and rhBMP-2/Bio-Gide composite membrane can promote the proliferation of osteoblasts and the expressions of osteogenic proteins, which has significant potential for bone tissue regeneration.
