摘  要:目的 探究马铃薯抗性淀粉(RS)对高脂血症大鼠肝脏固醇调节元件结合蛋白-1c(SREBP-1c)及其裂解激活蛋白(SCAP)以及乙酰辅酶A羧化酶1(ACC1)的影响。方法 将48只健康SPF级雄性SD大鼠随机分为空白对照组(8只)和模型对照组(40只)。空白对照组给予维持饲料,模型对照组给予高脂饲料,喂养2周后,根据TC水平,将模型对照组40只大鼠随机分成5组,分别为高脂模型组及1、2、4 g/kg马铃薯RS组及阳性对照(10 mg/kg辛伐他汀)组,每组8只。采用灌胃方式进行暴露,暴露容量为2 ml/kg,连续染毒6周。分别采用qRT-PCR法和免疫组织化学染色法检测肝脏组织SREBP、SCAP及ACC1 mRNA及蛋白的表达水平。结果 与空白对照组相比,高脂模型组大鼠肝脏组织中SREBP-1c、ACC1及SCAP mRNA的水平均升高,差异有统计学意义(P<0.05);与高脂模型组相比,各剂量马铃薯RS组和阳性对照组大鼠肝脏组织中SREBP-1c和SCAP及2、4 g/kg马铃薯RS组大鼠肝脏组织中ACC1 mRNA水平均降低,差异有统计学意义(P<0.05)。与空白对照组相比,高脂模型组大鼠肝脏组织中SREBP-1c、SCAP蛋白的水平均升高,差异有统计学意义(P<0.05);与高脂模型组相比,各剂量马铃薯RS组大鼠肝脏组织中SREBP-1c、ACC1和SCAP蛋白的水平均降低,差异有统计学意义(P<0.05);且随着马铃薯RS暴露剂量的升高,高脂大鼠肝脏组织中SREBP-1c、ACC1和SCAP蛋白的水平均呈下降趋势。结论 马铃薯RS可以下调大鼠肝脏组织SREBP、SCAP及ACC1的表达,为马铃薯抗性淀粉调脂作用机制的进一步研究提供了参考。Objective To understand the effects of potato resistant starch(RS)on sterol regulatory element-binding protein-1c(SREBP-1c),SREBPs-cleacage-activating protein(SCAP)and acetyl-CoA carboxylase 1(ACC1)in the liver of hyperlipidemia rats..Methods Totally 48 healthy SPF male SD rats were randomly divided into blank control group(8 rats)and model control group(40 rats).The blank control group was given maintenance diet,and the model control group was given high-fat diet.After 2 weeks of feeding,40 rats in the model control group were randomly divided into 5 groups according to TC level,namely high-fat model group,1,2 and 4 g/kg potato RS group and positive control group(10 mg/kg simvastatin)group,with 8 rats in each group.The rats were exposed by gavage with 2 ml/kg for 6 consecutive weeks.The expression levels of SREBP,SCAP and ACC1 mRNA and protein in the liver tissues were detected by qRT-PCR and immunohistochemical staining,respectively.Results Compared with the control group,the mRNA levels of SREBP-1c,ACC1 and SCAP in the liver tissues in high-fat model group significantly increased(P<0.05).Compared with high-fat model group,SREGBP-1C and SCAP in the liver tissues in potato RS group and positive control group,and ACC1 mRNA levels in the liver tissues of rats in 2 and 4 g/kg potato RS group decreased with significant differences(P<0.05).Compared with the control group,the levels of SREBP-1c and SCAP protein in liver tissue in high-fat model group increased with significant difference(P<0.05).Compared with high-fat model group,the protein levels of SREGBP-1C,ACC1 and SCAP in the liver tissue in potato RS group at all doses significantly decreased(P<0.05).with dose dependent manner.Conclusion Potato RS can down-regulate the expression of SREBP,SCAP and ACC1 in rat liver tissue,which provides a reference for further study on the mechanism of potato resistant starch regulating lipid metabolism.
