摘  要:将荷花花瓣粉末依次用不同极性溶剂提取,所得各提取物分别经柱层析或沉淀法得到主要组分,同浓度下比较它们对1,1-二苯基-2-三硝基苯肼(DPPH)自由基的清除作用,筛选出活性较好的组分。采用制备薄层层析法从中分离出单体,测试其对自由基的清除率与浓度的相关性,并鉴定活性较好单体的结构。结果表明,荷花花瓣无水乙醇提取物中的一个组分以及甲醇提取物中的一个组分清除自由基活性均较好,从中分离出的四个主要单体对自由基清除率均与浓度呈正相关性,在所测浓度范围内,其中一个单体清除自由基的活性与对照Vc相当,结构经鉴定为槲皮素。The powder of the petals of Nelumbo nucifera Gaertn. was extracted in order by the different polar solvents, the obtained extracts were divided into main parts respectively through column chromatography or precipitation method. Their effects on scavenging 2,2-diphenyl-1-picryl hydrazinyl (DPPH) radical were compared at the same concentration, the parts with better activity were screened out. In which the monomers were obtained by the method of preparative thin layer chromatography. The correlation between the free-radical clearance rates and their concentrations were measured, and the structures of the monomers with good scavenging activity were determined. The results showed that one part obtained from the ethanol extract and one part obtained from the methanol extract had better radical-scavenging activity than the other parts from these extracts. Four monomers were obtained from these two parts, and they also showed good dose-effect rela⁃ tionship with the concentrations, in which there was one monomer with the activity of clearing free radical equivalent to the control Vc within the measured concentration range, the structure had been identified as quercetin.
