摘 要:以猴耳环(Pithecellobium clypearia(Jack)Benth.)茎叶为试材,采用超声波辅助提取法提取总黄酮和没食子酸,在单因素试验的基础上,用响应面法对提取工艺进行优化。结果表明,猴耳环总黄酮提取工艺中各因素影响大小顺序为料液比>超声功率>超声时间>乙醇浓度,猴耳环总黄酮最佳提取工艺为料液比1∶90,乙醇浓度75%,超声功率170 W,超声时间40 min,此条件下总黄酮得率为2.29%(理论值2.37%);没食子酸提取工艺中各因素影响大小顺序为乙醇浓度>料液比>超声时间,没食子酸最佳提取工艺为料液比1∶70,乙醇浓度65%,超声时间60 min,此条件下没食子酸提取率为5.98%(理论值6.04%)。本研究构建的模型拟合效果好,试验值与拟合值高度相关,对猴耳环中总黄酮和没食子酸提取具有较强的指导性,可为其后续研究提供科学依据。Taking the stems and leaves of Pithecellobium clypearia(Jack)Benth.as experiment materials,the total flavonoids and gal-lic acid were extracted by ultrasonic assisted extraction method.On the basis of single factor experiment,the extraction process was op-timized by response surface methodology.The results showed that the order of influence of different factors on extraction of total flavo-noids from Pithecellobium clypearia(Jack)Benth.was ratio of material and liquid>ultrasonic power>ultrasound time>ethanol con-centration.The optimal extraction condition was ratio of material and liquid 1∶90,ethanol concentration 75%,ultrasonic power 170 W and ultrasound time 40 min.Under this condition,the yield of total flavonoids was 2.29%(theoretical value 2.37%).The order of in-fluencing factors on gallic acid extraction was ethanol concentration>ratio of material and liquid>ultrasound time.The optimal extrac-tion condition was ratio of material and liquid 1∶70,ethanol concentration 65%and ultrasonic time 60 min.Under this condition,the extraction rate of gallic acid was 5.98%(theoretical value 6.04%).The model established in this study had a good fitting effect,and the experimental values were highly correlated with the fitting values,which had strong guidance for the extraction of total flavonoids and gallic acid from Pithecellobium clypearia(Jack)Benth.,and could provide a scientific basis for the subsequent research.
