摘 要:本研究旨在建立利用全自动免疫磁珠净化结合高效液相色谱测定饲料中脱氧雪腐镰刀菌烯醇(呕吐毒素)的方法。饲料样品用水提取,经全自动免疫磁珠净化后,采用高效液相色谱仪测定,外标法定量。结果表明:在浓度为0.05~2.0μg/mL呕吐毒素的线性关系良好(R^(2)=0.9999),方法的检出限为0.03 mg/kg,定量限为0.1 mg/kg;呕吐毒素在3个添加浓度(0.1、0.2、1.0 mg/kg)水平下的平均回收率为85.7%~102.5%,相对标准偏差为2.2%~9.6%。与传统固相萃取柱、免疫亲和柱净化相比,免疫磁珠净化集提取、分离、净化、浓缩于一体,可实现完全自动化处理,显著提高工作效率,避免因人为操作导致的结果偏差,适用于大批量饲料样本中呕吐毒素的测定。This study aimed to establish an analytical method for the determination of deoxynivalenol(DON)in feeds using automatic immunomagnetic beads clean-up combined with high-performance liquid chromatography.Feed samples were extracted with water,purified by automatic immunomagnetic beads,and then detected by high-performance liquid chromatography,with external standard method for quantification.The results indicated that good linearity was obtained for DON in the concentration ranges of 0.05~2.0μg/mL with the corresponding correlation coefficients(R^(2))higher than 0.9995.The LOD and LOQ of the proposed method were 0.03 mg/kg and 0.1 mg/kg,respectively.The average recoveries of the fortified samples(0.1,0.2 mg/kg and 1.0 mg/kg)were 85.7%~102.5%,with the relative standard deviations(RSD)in the ranges of 2.2%~9.6%.Compared with the traditional solid-phase extraction and immunoaffinity column purification methods,immunomagnetic beads technique integrates the extraction,separation and purification in a single run.It could achieve fully automated processing,which significantly improving the work efficiency and avoiding result deviations caused by manual operation.Thus,it was much more suitable for the determination of DON in a large-scale number of feed sample.
