摘 要:为挖掘传统发酵乳制品中具有潜在益生特性的开菲尔慢生乳杆菌(Lentilactobacillus kefiri,L.kefiri)菌株。文章对10株L.kefiri进行体外益生特性检测,结果表明,L.kefiri NM119-2相较于其他菌株,益生性能良好,该菌株在pH=3的模拟人工胃液中3 h存活率为57.55%,在0.3%胆盐中4 h存活率为95.79%;对氯霉素、青霉素、环丙沙星和庆大霉素等6种抗生素显示敏感;疏水性达80.36%;静置24 h后自凝聚能力为43.30%;DPPH自由基清除率为97.91%,羟自由基清除率为52.2%,超氧阴离子清除率为27.81%,综合抗氧化能力强。碳水化合物利用能力结果显示,L.kefiri NM119-2可利用D-葡萄糖、D-乳糖和D-果糖等9种糖。全基因组测序结果显示,L.kefiri NM119-2基因组全长为2.48 Mb,编码2611个基因,涵盖了氨基酸运输和代谢、碳水化合物代谢和磷酸化作用等相关基因;L.kefiri NM119-2耐药基因均位于染色体上,安全风险较低。To exploit the potential probiotic properties of Lentilactobacillus(L.)kefiri in traditional fermented dairy products.The probiotic properties of 10 strains of L.kefiri were tested in vitro,the results indicated that L.kefiri NM119-2 possessed superior probiotic properties compared to the other strains,with a 3 h survival rate of 57.55%,in artificial gastric juice at pH=3 and 95.79%in bile salts at 0.3%for 4 h.L.kefiri NM119-2 exhibited sensitivity to six antibiotics,including chloramphenicol,penicillin,ciprofloxacin and gentamicin.And it had a hydrophobicity of 80.36%,the auto-aggregation capacity was 43.30%after 24 h of standing,and it possessed strong antioxidant capacity,among them,the scavenging rate of DPPH was 97.91%,the scavenging rate of hydroxyl radicals was 52.2%,and the scavenging rate of superoxide anion was 27.81%.The results of carbohydrate utilization showed that L.kefiri NM119-2 could utilize nine types of sugars,including D-glucose,D-lactose and D-Fructose.Whole genome sequencing of L.kefiri NM119-2 was demonstrated.The whole genome sequencing results showed the total length of L.kefiri NM119-2 was 2.48 Mb,encoding 2611 genes,including genes related to amino acid transport and metabolism,carbohydrae metabolism and phosphorylation.The resistance genes of L.kefiri NM119-2 were all located on chromosomes,which is a low safety risk.
