基于网络药理学探讨中药防治结直肠癌的研究进展

摘  要:结直肠癌的发病率和病死率都居癌症前列,给人类健康造成严重损害。结直肠癌早期发病隐匿,多数患者发现患病时已失去手术机会。目前,化疗仍是该病主要的治疗手段,但化疗具有全身毒性,患者接受治疗时常出现恶心、呕吐及骨髓抑制的情况,且易耐药,影响继续治疗。近年来中医药治疗结直肠癌受到广大学者的密切关注。临床实践表明,中药防治结直肠癌疗效显著,但治疗机制不明确一直影响其应用与发展。网络药理学的出现为中药治疗结直肠癌分子机制的研究打开了一扇新大门,丰富的生物信息分析数据和分子对接数据能够对其活性成分、治疗通路及作用靶点进行预测,直观地呈现三者之间的关系。此外,基于网络药理学探讨中药治疗机制的研究层出不穷,为疾病的临床治疗及机制研究提供了新方向。本研究基于网络药理学对近年来中药单体化合物、中药单药、中药药对、中药复方防治结直肠癌的研究进行综述,发现治疗结直肠癌的主要活性成分有槲皮素、山柰酚、β–谷甾醇、豆甾醇、木犀草素、柚皮素等,常见的治疗靶点有丝氨酸–苏氨酸蛋白激酶1(Akt Serine/Threonine Kinase 1,AKT1)、肿瘤蛋白p53(Tumor Protein p53,TP53)、Kirsten大鼠肉瘤病毒癌基因同源物、B淋巴细胞瘤–2(B–cell Lymphoma–2,Bcl–2)、表皮生长因子受体(Epidermal Growth Factor Receptor,EGFR)、肿瘤坏死因子(Tumor Necrosis Factor,TNF)、雌激素受体1(Estrogen Receptor 1,ESR1)、白细胞介素(Interleukin,IL)–6等,癌症信号通路、促炎癌变机制和细胞凋亡等则为常见的治疗途径。总之,中药可通过调整细胞周期、抗炎、促进凋亡等多种生物途径及多靶点、多通路参与结直肠癌的治疗。The incidence and mortality of colorectal cancer are in the forefront of cancer,causing serious damage to human health.The early onset of colorectal cancer is occult,and most patients have lost the opportunity of surgery when they are found to have the disease.At present,chemotherapy is still the main treatment method for this disease.However,chemotherapy has systemic toxicity,patients often suffer from nausea,vomiting and bone marrow suppression during treatment,and drug resistance is easy to occur,affecting the continuation of the treatment.In recent years,the treatment of colorectal cancer in TCM has been paid close attention by many scholars.Clinical practice has shown that TCM is effective in the prevention and treatment of colorectal cancer,but the unclear action mechanism has always been a problem in its application and development.The emergence of network pharmacology has opened a new door for the study on the molecular mechanism of TCM on colorectal carcer.Abundant biological information analysis data and molecular docking data can predict its active ingredients,therapeutic pathways and action targets,and visually present the relationships among the three.In addition,the research on the action mechanism of TCM based on network pharmacology emerge in an endless stream,providing a new direction for the clinical treatment and mechanism research of diseases.Based on network pharmacology,the studies on the prevention and treatment of colorectal cancer with TCM monomer compound,TCM single medicine,TCM couplet medicines and TCM compound were reviewed in this study.It was found that quercetin,kaempferol,β-sitosterol,stigmasterol,luteolin,and naringin were the main active ingredients commonly used in the treatment of colorectal cancer.The common therapeutic targets included AKT1,TP53,KRAS,Bcl-2,EGFR,TNF,ESR1,IL-6,etc.Cancer pathway,proinflammatory carcinogenesis mechanism and apoptosis were common therapeutic pathways.All in all,Chinese medicine can be involved in the treatment of colorectal cancer by regulating
