摘 要:为探究樱桃李多酚提取物(P.cerasifera polyphenol extract,PPE)对非酒精性脂肪肝(non-alcoholic fatty liver disease,NAFLD)小鼠肝脏的保护作用及其潜在的保肝机制,以PPE干预高脂饮食诱导的NAFLD小鼠,对其血清生化指标、肝脏病理变化及肝脏非靶向代谢组进行分析。结果表明,PPE能显著降低NAFLD小鼠血清甘油三酯(triglycerides,TG)、总胆固醇(total-cholesterol,TC)、谷草转氨酶(aspartate aminotransferase,AST)和谷丙转氨酶(alanine aminotransferase,ALT)含量及肝脏小叶内炎症、脂肪变性和气球样变性。PPE能逆转NAFLD小鼠肝脏代谢物变化,并通过增强机体抗氧化活性来缓解NAFLD,从对照(CON,灌胃0.3 mL/d生理盐水)组、模型(HFD,灌胃0.3 mL/d生理盐水)组和多酚给药[HFD+H,灌胃400 mg/(kg·d)PPE,0.3 mL]组中共筛选出41个差异代谢物,主要富集于嘧啶代谢、蛋白质消化和吸收、代谢途径、甘氨酸、丝氨酸和苏氨酸代谢和ABC转运蛋白等通路中。This study aimed to explore the protective effect of P.cerasifera polyphenol extract(PPE)on the liver of mice with non-alcoholic fatty liver disease(NAFLD)and its potential hepatoprotective mechanism.PPE was utilized to intervene in high-fat diet-induced NAFLD mice,and the serum biochemical indicators,liver pathological changes,and liver non-targeted metabolome of the mice were analyzed.The results showed that PPE could significantly reduce serum triglycerides(TG),total-cholesterol(TC),aspartate aminotransfer-ase(AST),and alanine aminotransferase(ALT),content as well as inflammation,steatosis,and ballooning de-generation in liver lobules in mice with NAFLD.PPE could reverse the changes in liver metabolites in NAFLD mice and alleviate NAFLD by enhancing the body′s antioxidant activity.A total of 41 differential metabolites were screened out from the CON(control,intragastric administration of 0.3 mL/d saline)group,HFD(high fat diet,intragastric administration of 0.3 mL/d saline)group,and HFD+H[high fat diet and high-dose PPE,intra-gastric administration of 0.3 mL of 400 mg/(kg·d)PPE]group,mainly enriching in pyrimidine metabolism,protein digestion and absorption,metabolic pathways,glycine,serine and threonine metabolism,ABC transport-ers,and other pathways.
