摘 要:为优化巴戟天多糖提取工艺,并考察其体外抗氧化活性,以单因素试验(料液比、酶解时间、酶量、超声时间为因素)、超声辅助酶法提取试验和正交试验探索其工艺条件。同时,采用DPPH法、超氧阴离子法、ABTS法和总抗氧化能力法评价巴戟天多糖的抗氧化能力。结果表明,相较于单独酶法和超声法,联合法有利于巴戟天多糖的提取。最优工艺条件为料液比1∶40 g/mL、纤维素酶量2500 U/g、酶解时间1.5 h、超声时间40 min,先酶解处理后超声提取。在此条件下,巴戟天多糖得率达8.55%±0.05%。巴戟天多糖提取物有一定的抗氧化活性,尤其具有良好的ABTS自由基清除和总抗氧化能力,为后续相关食药用资源开发提供一定依据。To optimize the extraction process and to investigate its antioxidant activity in vitro of polysaccharides from Morinda officinalis,the single-factor experiment(with solid-liquid ratio,enzymolysis time,enzyme amount,and ultrasonic time as factors),ultrasonic-assisted enzymolysis extraction test and orthogonal test were designed to explore the conditions.At the same time,the antioxidant capacity of polysaccharides was evaluated using DPPH,superoxide anion,ABTS,and total antioxidant capacity methods.The results showed that the combined method was favorable for extracting polysaccharides compared with the separate enzymatic and ultrasonic techniques.The optimal conditions were as follows:solid-liquid ratio 1:40 g/mL,amount of cellulase 2500 U/g,enzymatic hydrolysis time 1.5 h,ultrasonic time 40 min,enzymolysis followed by ultrasonication.Under this condition,the extraction rate of polysaccharides reached 8.55%±0.05%.Morinda officinalis polysaccharides showed certain antioxidant activity,especially in scavenging ABTS free radical and total antioxidant capacity,and provided the basis for the subsequent development of related food and medicinal resources.
