杠板归抑菌活性部位的筛选及其作用机理初步研究

摘  要:筛选杠板归不同提取部位的抑菌活性,初步探讨活性部位的作用机理。采用牛津杯法和两倍稀释法测定提取物体外抑菌活性,测定菌液中的蛋白质含量、碱性磷酸酶(AKP)含量,采用十二烷基硫酸钠-聚丙烯酰胺凝胶电泳法对细菌蛋白合成的分析,采用荧光分光光谱法对细菌基因组DNA的分析,综合探索抑菌机理。试验结果表明,杠板归提取物B和F对金黄色葡萄球菌和大肠杆菌均有良好的抑菌作用,提取物B质量浓度10 mg/mL时对金黄色葡萄球菌和大肠杆菌的抑制效果明显,其抑菌圈分别为22.67 mm和21.33 mm,MIC均为3.125 mg/mL。随着提取物B对指标菌作用时间增加,菌液中可溶性蛋白质含量增加和碱性磷酸酶含量增加,表明菌体中2类物质已泄漏。荧光光谱显示提取物B能使指标菌基因组DNA荧光淬灭,表明提取物B能破坏大肠杆菌和金黄色葡萄球菌基因组DNA结构。杠板归提取物B对大肠杆菌和金黄色葡萄球菌具有良好的抑菌作用,其作用机理可能与破坏细胞壁、细胞膜和基因组DNA结构有关。Screen antibacterial active parts of different solvent extracts of Polygonum perfoliatum L.and preliminary study on its mechanism of action.The antibacterial activity of the extracts was evaluated by Oxford cup method and the double dilution method in vitro.Through the amount of alkaline phosphatase and protein in bacterial solution,SDS-PAGE gel electrophoresis analysis of protein synthesis,and fluorescence spectroscopic analysis of genomi c DNA,the mechanism of antibacterial was explored.In the experiments,extract B and extract F had the inhibitory effects on Staphylococcus Aureus and Escherichia Coli,extract B with concentrations 10 mg/mL was the strongest inhibitory effect of Staphylococcus aureus and Escherichia coli,the inhibitory zone was 22.67 mm and 21.33 mm,respectively,and MIC was 3.125 mg/mL.The amount of alkaline phosphatase and protein in bacterial solution significantly increased with the prolongation of time.It indicated that the bacterial alkaline phosphatase and protein leakage.Fluorescence spectroscopic analysis indicated that extract B could lead to fluorescence quenching of genomic DNA,suggesting that extract B could damage genomic DNA.The extract B from Polygonum perfoliatum L.showed good antibacterial activity against Staphylococcus aureus and Escherichia coli,and its mechanism might related to the cell wall and cell membrane was destroyed,and disrupting genomic DNA.
